Abstract An ultrasensitive ICP‐MS aptasensor is developed utilizing a label‐free, simple filter membrane‐assisted separation technique combined with nucleic acid signal amplification for the analysis of circulating tumor cells (CTCs) in lung cancer clinical samples. The approach based on high‐affinity interaction between aptamers and PD‐L1 mucin 1, which are overexpressed cell surface, conjunction functional Y‐DNA nanospheres catalytic hairpin assembly amplifications, enabling simultaneous detection two proteins. Additionally, four‐armed nanostructure significant spatial site resistance self‐assembled by introducing streptavidin biotinylated‐hairpin structures, improving efficiency membrane. This structural design enables effective isolation biotin‐T‐Hg 2+ ‐T biotin‐C‐Ag + ‐C from free Hg Ag , facilitating highly sensitive dual‐protein via ICP‐MS. limits reached ag mL −1 levels proteins single‐cell A549 cells. Herein, a supramolecular DNA nanodevice was formed via the rolling circle amplification (RCA) and hybridization chain reaction (HCR) cascade on tetrahedral nanostructure (TDN) to achieve simultaneous sensitive detection intracellular imaging of dual-miRNAs related liver cancer. The effectively addressed limitations low probe loading capacity in traditional TDN nanodevices by enriching plenty signal probes around single TDN, significantly enhancing fluorescence signal. Impressively, with fulcrum dense structure imparted strong rigidity, ensuring stability decrease aggregation quenching for further increasing response. Consequently, biosensor based enabled miRNA221 miRNA222, achieved accurate situ displaying improved capabilities compared TDN-based nanodevices. More importantly, precise miRNA222 could distinguish hepatocellular carcinoma cells different degrees metastasis from human normal cells, providing more information diagnosis development carcinoma.

Fluorescence sensing is widely used in vitro detection due to its high sensitivity and rapid result delivery. However, systems based on nanomaterials involving complex cumbersome purification steps can lead sample loss significantly reduce the accuracy of results. To address this issue, we proposed a lanthanide-based aptasensor featuring target-triggered antenna effect enhance time-resolved luminescence (TRL) chelated Tb3+ combined with wash-free strategy. This sensor enabled highly sensitive epithelial cell adhesion molecule (EpCAM) exosomes from tumors, eliminating necessity for thereby delivering more stable reliable Specifically, using computer-aided design, sequence capable self-folding into hairpin structure (Antenna-Hairpin-Tb) was obtained. After hybridization an EpCAM-specific aptamer (AptEpCAM), duplex (Probe-Tb) constructed detection. When targeted binding occurred between AptEpCAM exosomes, Antenna-Hairpin-Tb dehybridized Probe-Tb underwent self-folding, leading significant sensitized TRL signal effect-induced energy transfer ligand Tb3+. Over broad range exosome concentrations (spanning 6.19 × 102 108 particles/mL), emission intensity at 543 nm linearly increased concentration (linear relationship: Y = 332.62 lg(Nexosomes) + 3690.5, R2 0.9956), achieving theoretical limit as low 17 particles/mL buffer. Furthermore, swift effective differentiation all cancer serum samples nonepithelial ones within just 30 min operation, 100% sensitivity, specificity, accuracy.
