The concept of dedifferentiation and reprogramming mature somatic cells holds much promise for the three-front "war" against tissue damage, cancer, aging. It was hoped that human into induced pluripotent state, along with use embryonic stem cells, would transform regenerative medicine. However, despite global efforts, clinical applications remain a distant dream, due to associated factors such as genomic instability, tumorigenicity, immunogenicity, heterogeneity. Meanwhile, expression (pluripotent) markers in multiple cancers has baffled scientific community, it been suggested dedifferentiate "reprogram" state vivo initiate cancer. also aging can be reversed by partial vivo. better methods are needed; using vectors or Yamanaka vivo, example, is dangerous, many potential anti-aging therapies carry same risks those described above. present perspective examines endogenous, very small embryonic-like (VSELs). These naturally tissues; they routinely replace diseased ensure regeneration maintain life-long homeostasis, have ability differentiate adult counterparts. Recent evidence suggests selective expansion epigenetically altered VSELs their blocked differentiation. Furthermore, VSEL numbers directly linked lifespan studies long- short-lived transgenic mice, dysfunction found ovaries aged mice. To conclude, greater interest VSELs, address all three fronts this war, could "light at end tunnel."

As women age, their ovarian follicle pool naturally declines. However, female germline stem cells (FGSCs) possess a unique ability to differentiate into oocytes and continuously self-renew, providing an effective means of delaying aging by replenishing the primordial pool. Therefore, activating FGSCs is critical in reshaping safeguarding function. In this study, we investigated biological activity proanthocyanidins (PACs), natural antioxidants that exhibit anti-aging anti-inflammatory properties beneficial for both male reproduction. Our vivo vitro experiments demonstrate PACs promote proliferation while aging. increase number follicles, primary corpus luteum reducing cystic elevate estradiol (E2) levels along with anti-mullerian hormone (AMH) concentration mice. Additionally, significantly boost time- dose-dependently upregulating mRNA & protein expressions FGSCs-specific markers such as MVH OCT4 downregulating p53/p21 via activation silent information regulator 1(Sirt1) signaling pathway. The effects on FGCSs were found be impeded Sirt1 inhibitor EX527. PACS delay premature insufficiency (POI) through regulating Sirt1-p53-p21 pathway involving FGSCs.

Abstract Autoimmune factors play an important role in premature ovarian insufficiency (POI). Human amniotic epithelial stem cells (hAESCs) have recently shown promising treatment effects on chemotherapy-induced POI. However, the therapeutic efficacy and underlying mechanisms of hAESCs autoimmune POI remain to be investigated. In this study, we showed for first time that intravenous transplantation could reside ovary zona pellucida 3 peptide (pZP3) induced mice model at least 4 weeks. improve function fertility, alleviate inflammation reduce apoptosis granulosa (GCs) mice. The transcriptome analysis ovaries vitro co-cultivation experiments suggest activation AKT ERK pathways may key mechanism effect hAESCs. Our work provides theoretical experimental foundation optimizing administration hAESCs, as well clinical application patients. Graphical

There is a group of polycystic ovary syndrome (PCOS) patients in clinic who have diminished ovarian reserve (DOR) combination. This study was designed to evaluate the differences glucolipid metabolism, hypothalamic-pituitary-ovarian (HPO) axis-related parameters, and autoimmune antibodies PCOS with without DOR.A total 2307 patients, including 1757 alone 550 both DOR, were enrolled this retrospective study. Parameters HPO measured analyzed. The prevalence DOR among all 23.84%. Many such as follicle-stimulating hormone (FSH), luteinizing (LH), estradiol (E2), prolactin (PRL) significantly different compared DOR. The FSH levels positively correlated LH, testosterone (T), androstenedione (AD) levels, but had no association metabolism after adjusting for body mass index (BMI). Moreover, anti-ovarian antibody (AOAb) anti-21-OH (21-OHAb) elevated DOR.PCOS showed more chaotic axis antibodies, suggesting that factors may be cause women PCOS.
