DNA walking machines have achieved significant breakthroughs in areas such as biosensing, bioimaging, and early cancer diagnosis, facilitated by the self-assembly of or its combination with other materials, magnetic beads metal nanoparticles. However, current machine strategies are constantly challenged inadequate analytical sensitivity, while sophisticated signal amplification procedures often indispensable. Single-particle inductively coupled plasma mass spectrometry (SP-ICPMS) provides superior sensitivity can effectively discriminate between background noise detected signals due to large number atoms a nanoparticle concentrating effect single detection. In this study, we present novel approach utilizing counting duplex-specific nuclease (DSN)-assisted construct 3D for detecting aggressive prostate (PCa) biomarker miRNA-200c. Traditional microRNA detection methods have some limitations that cannot be ignored. Enzyme-catalyzed nucleic acid amplification has gradually become an important method in bioanalysis and is expected to a substitute for traditional methods. Combining enzymes with different functions provides wide range of analytical design possibilities create high-performance sensing strategies. Meanwhile, the optical tweezers produce tightly focused laser beam are integrated suspension bead array fully concentrates signal labels, contributing stable output improved efficiency. Herein, we developed novel "DSN-RCA dual tweezer-assisted imaging" strategy highly efficient microRNA-21 (miRNA-21). The conversion low abundance targets into bulk fluorescence probes was achieved by duplex-specific nuclease (DSN), enrichment on supports rather than dispersion solution realized rolling circle (RCA) one step. synergistic integration application effectively sensitivity specificity. Finally, created modified tweezer system, which high-quality imaging analysis proposed limit (LOD) as 5.90 pM excellent specificity, stability, reproducibility successfully applied human serum samples, great potential clinical serological research.
