More than 5 years have passed since the Diffusion Tensor Image Analysis ALong Perivascular Space (DTI-ALPS) method was proposed with intention of evaluating glymphatic system. This is handy due to its noninvasiveness, provision a simple index in straightforward formula, and possibility retrospective analysis. Therefore, ALPS adopted evaluate system for many disorders studies. The purpose this review look back discuss at moment.

Neurodegenerative diseases (ND) are characterized by the accumulation of aggregated proteins. The glymphatic system, through its rapid exchange mechanisms between cerebrospinal fluid (CSF) and interstitial (ISF), facilitates movement metabolic substances within brain, serving functions akin to those peripheral lymphatic system. This emerging waste clearance mechanism offers a novel perspective on removal pathological in ND. article elucidates recent discoveries regarding system updates relevant concepts model. It discusses potential roles ND, including Alzheimer's disease (AD), Parkinson's (PD), multiple atrophy (MSA), proposes as therapeutic target for these conditions.

The aim of the study was to evaluate glymphatic function and its related factors in patients with Parkinson's disease (PD) PD different ages using diffusion tensor image analysis along perivascular space (DTI-ALPS) method.Medical records imaging data 93 idiopathic 42 age- sex-matched healthy controls (HCs) were retrospectively reviewed analyzed. diffusivity spaces, projection fibers, association fibers calculated on (DTI) acquire (ALPS) index.PD exhibited a reduced ALPS index compared HCs. Negative correlations between clinical information including age, age at onset, sleep scale 2nd version (PDSS-2) scores, history diabetes mellitus revealed group. Besides, negative correlation severity motor symptoms identified subgroup aged 65 above, rather than younger ones. The results demonstrate that index, potential noninvasive measure compromised activity, is involved pathophysiology PD, especially ones those disorders.

Abstract Recently, an astrocytic aquaporin 4-dependent drainage system, that is, the glymphatic has been identified in live murine and human brain. Growing evidence suggests function is impaired patients with several neurodegenerative diseases, including Alzheimer’s Parkinson’s disease. As third most common disease, although animal studies have indicated early dysfunction likely important pathological mechanism underpinning amyotrophic lateral sclerosis (ALS), no available study conducted to thoroughly assess vivo ALS date, particularly early-stage ALS. Thus, using diffusion tensor imaging analysis along perivascular space (ALPS) index, approximate measure of vivo, we aimed explore whether ALS, diagnostic performance ALPS index distinguishing between healthy subjects. We also identify relationships clinical disabilities sleep problems In this retrospective study, King’s Stage 1 were defined as enrolled 56 32 age- sex-matched control All participants completed screening, assessment analysis. For assessment, Pittsburgh Sleep Quality Index, Epworth Sleepiness Scale polysomnography used. Compared subjects, had a significantly lower after family-wise error correction (P < 0.05). Moreover, receiver operating characteristic showed area under curve for was 0.792 (95% confidence interval 0.700–0.884). Partial correlation analyses correlated disability disturbances Multivariate efficiency (r = 0.419, P 0.002) periodic limb movements −0.294, 0.017) significant predictive factors conclusion, our continues support role pathology, provide additional insights into value its suggest improvement may be promising strategy slowing process Future are needed therapeutic individuals presymptomatic-stage diseases.

Emerging pathological evidence suggests that there is an association between glymphatic dysfunction and the progression of Parkinson's disease (PD). However, clinical this remains lacking. In study, index for diffusion tensor image analysis along perivascular space (ALPS index) was calculated to evaluate function.Overall, 289 patients with PD were enrolled in cross-sectional study. The ALPS found be negatively correlated age, severity, dyskinesia. In longitudinal information on a total 95 5-year follow-up examinations collected from Progression Marker Initiative, 33 which classified into low group, all others mid-high group based first tertile baseline index. results regression indicated significant main effect autonomic dysfunction, as well activities daily living. addition, had faster deterioration MDS-UPDRS part III II, Symbol Digit Modalities Test Hopkins Verbal Learning Test. Abstract Cerebrospinal fluid (CSF) plays a critical role in metabolic waste clearance from the brain, requiring its circulation throughout various brain pathways, including ventricular system, subarachnoid spaces, para‐arterial interstitial and para‐venous spaces. The complexity of CSF has posed challenge obtaining noninvasive measurements dynamics. assessment dynamics circulatory pathways is possible using diffusion magnetic resonance imaging (MRI) with optimized sensitivity to incoherent water movement across brain. Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are fatal neurodegenerative disorders often co-occurring in the same patient, a feature that suggests common origin of two diseases. Consistently, pathological inclusions proteins as well mutations genes can be identified both ALS/FTD. Although many studies have described several disrupted pathways within neurons, glial cells also regarded crucial pathogenetic contributors Here, we focus our attention on astrocytes, heterogenous population perform functions for optimal central nervous system homeostasis. Firstly, discuss how post-mortem material from ALS/FTD patients supports astrocyte dysfunction around three pillars: neuroinflammation, abnormal protein aggregation, atrophy/degeneration. Furthermore, summarize current attempts at monitoring living using either novel imaging strategies or soluble biomarkers. We then address pathology is recapitulated animal cellular models used these to understand molecular mechanisms driving platforms pre-clinical testing therapeutics. Finally, present clinical trials ALS/FTD, restricting discussion treatments modulate functions, directly indirectly.
