Tumor-specific, circulating cell-free DNA in liquid biopsies is a promising source of biomarkers for minimally invasive serial monitoring treatment responses cancer management. We will review the current understanding origin and different forms release (including various types cell death active secretion processes) clearance routes. The dynamics extracellular blood during therapy role pathophysiological processes (tumor-associated inflammation, NETosis, pre-metastatic niche development) provide insights into mechanisms that contribute to tumor development metastases formation. Better knowledge tumor-specific could facilitate new therapeutic diagnostic options

Despite being the hallmark of cancer that is responsible for highest number deaths, very little known about biology metastasis. Metastatic disease typically manifests after a protracted period undetectable following surgery or systemic therapy, owing to relapse recurrence. In case breast cancer, metastatic can occur months decades initial diagnosis and treatment. this review, we provide an overview key factors influence recurrence, with goal highlighting critical unanswered questions still need be addressed make difference in mortality patients.

Lung metastasis is the major cause of breast cancer-related mortality. The neutrophil-associated inflammatory microenvironment aids tumor cells in metastatic colonization lungs. Here, we show that tumor-secreted protease cathepsin C (CTSC) promotes breast-to-lung by regulating recruitment neutrophils and formation neutrophil extracellular traps (NETs). CTSC enzymatically activates membrane-bound proteinase 3 (PR3) to facilitate interleukin-1β (IL-1β) processing nuclear factor κB activation, thus upregulating IL-6 CCL3 for recruitment. In addition, CTSC-PR3-IL-1β axis induces reactive oxygen species production NETs, which degrade thrombospondin-1 support growth cancer expression secretion are associated with NET lung human tumors. Importantly, targeting compound AZD7986 effectively suppresses a mouse model. Overall, our findings reveal mechanism how regulate niches CTSC-targeting approaches treatment.

Abstract Metabolic rewiring and epigenetic remodeling, which are closely linked reciprocally regulate each other, among the well-known cancer hallmarks. Recent evidence suggests that many metabolites serve as substrates or cofactors of chromatin-modifying enzymes a consequence translocation spatial regionalization metabolites. Various metabolic alterations modifications also reportedly drive immune escape impede immunosurveillance within certain contexts, playing important roles in tumor progression. In this review, we focus on how reprogramming cells reshapes alterations, particular acetylation methylation histone proteins DNA. We discuss other eminent such as, succinylation, hydroxybutyrylation, lactylation, update current advances metabolism- modification-based therapeutic prospects cancer.
