Abstract Glycosylation, a key mode of protein modification in living organisms, is critical regulating various biological functions by influencing folding, transportation, and localization. Changes glycosylation patterns are significant feature cancer, associated with range pathological activities cancer‐related processes, serve as biomarkers providing new targets for cancer diagnosis treatment. Glycoproteins like human epidermal growth factor receptor 2 (HER2) breast alpha‐fetoprotein (AFP) liver carcinoembryonic antigen (CEA) colon prostate‐specific (PSA) prostate all tumor approved clinical use. Here, we introduce the diversity structures newly discovered substrate—glycosylated RNA (glycoRNA). This article focuses primarily on metastasis, immune evasion, metabolic reprogramming, aberrant ferroptosis responses, cellular senescence to illustrate role cancer. Additionally, summarize applications diagnostics, treatment, multidrug resistance. We envision promising future glycosylation.

Metabolic changes are coupled with alteration in protein glycosylation. In this review, we will focus on macrophages that pivotal the pathogenesis of pulmonary fibrosis and sarcoidosis thanks to their adaptable metabolism an attractive therapeutic target. Examples presented review demonstrate glycosylation regulates metabolism-driven immune responses macrophages, implications for fibrotic processes granuloma formation. Targeting proteins regulate glycosylation, such as fucosyltransferases, neuraminidase 1 chitinase could effectively block immunometabolic driving inflammation fibrosis, providing novel avenues interventions.

The role of peripheral blood platelets as indicators cancer progression is increasingly recognized, and the significance abnormal glycosylation in platelet function related disorders gaining attention. However, potential a source protein site-specific for diagnosis remains underexplored. In this study, we proposed general pipeline that integrates quantitative proteomics with glycoproteomics, allowing an in-depth investigation glycoproteome. With pipeline, generated data set comprising 3,466 proteins qualitative information, 3,199 3,419 glycans information 3,377 from hepatocellular carcinoma (HCC) patients, metastatic liver (mLC) healthy controls. integrated analysis revealed significant changes N-glycosylation patients. Further systems biology lectin pull-down-coupled ELISA assays independent clinical samples confirmed two N-glycoproteins specific glycan types, complement C3 (C3) oligomannose modification integrin β-3 (ITGB3) sialylation, biomarkers distinguishing patients individuals, without differentiating between HCC mLC patient group. These findings highlight biomarkers.

COPD is a multifactorial illness characterized by long-term restriction of airflow and an inflammatory reaction in the lungs. The associated emphysema leads to breakdown alveolar proteins abnormal expansion lung air spaces. Chronic bronchitis caused same disease can result increased deposition structural proteins, narrowing airways, excessive mucus secretion leading acute exacerbation (AECOPD). most commonly prescribed medications for it, such as glucocorticoids bronchodilators, provide important therapeutic benefits, but they also have negative side effects, including immunosuppression infection. Therefore, it necessary develop treatment that specifically target immune system molecular components. This review focuses on non-eosinophilic aspects immunological modulation management. Since, existing literature extensively covers eosinophilic inflammation, this aims fill gap examining alternative pathways their implications. findings suggest targeting specific responses may enhance efficacy while minimizing adverse effects with traditional therapies. In summary, emphasizes importance advancing research into mechanisms COPD, prescribing development novel therapies more effectively manage disease.
