Over the past two decades, whole food supplementation strategies have been leveraged to target mental health. Abstract Dietary intake is tightly coupled to gut microbiota composition, human metabolism, and the incidence of virtually all major chronic diseases. nutrient are usually quantified using dietary questionnaires, which tend focus on broad food categories, suffer from self-reporting biases, require strong compliance study participants. Here, we present MEDI (Metagenomic Estimation Intake): a method for quantifying food-derived DNA in stool metagenomes. Abstract Objective We assessed the impact of a food‐provisioning intervention on diet quality in children with obesity. Methods Participants ( n = 33, aged 6–11 years) were randomly assigned to either usual care (intensive health behavior and lifestyle treatment) or (usual + food provisioning; high‐fiber, low‐dairy diet) for 4 weeks. The primary outcome was change child at Week 4. Secondary outcomes changes weight, insecurity, gut microbiome composition (16S ribosomal RNA), dietary intake, measured via an objective DNA‐based biomarker (i.e., FoodSeq). Genomic data analyzed against larger pediatric adolescent obesity cohort 195, 10–18 from similar households. Results Intervention demonstrated across all components more effective than increasing whole grain (β 0.20, 95% CI: 0.05 0.34; p 0.013) fiber 2.52, 1.28 3.76; < 0.001) decreasing dairy −1.31, −2.02 −0.60; 0.001). FoodSeq results, highly concordant grocery orders (adjusted R 2 0.65; 0.001), indicated shift toward low‐energy‐density plant taxa relative prior survey related 8.64, 5.18 12.14; No significant observed microbiome, insecurity. Abstract Between birth and adulthood, the human gut is colonized by a complex microbial community. Despite established links between infant microbiome health, knowledge limited for how complementary feeding influences colonization. Using FoodSeq, an objective DNA-based dietary assessment technique, we analyzed 1,036 fecal samples from 729 children aged 0-3 years across countries in North America, Central Africa, Asia. We detected wide diversity of 199 unique plant food sequences, which only eight staple foods were consistently present all countries. this variation global diet, identified universal trajectories early life exposure: weaning stage, tracked with diversity, emerged as dominant signature populations. Still, did not correlate diversity. Instead, stage specifically predicted abundance adult-like bacterial taxa, including known fiber-degrading after age 1. Our findings support two-stage model maturation: phase dominated milk-adapted taxa independent feeding, followed maturation where diet shapes microbiota This suggests that tracking promoting may timely emergence microbiome.
