Maternal vaccinations administered prior to conception or during pregnancy enhance the immune protection of newborn infants against many pathogens. A feasibility experiment was conducted determine if monkeys can be used model placental transfer maternal antibody SARS-CoV-2. Six adult rhesus were immunized with adjuvanted recombinant-protein antigens comprised receptor-binding domain human IgG1-Fc fusion proteins (RBD-Fc) containing protein sequences from ancestral-Wuhan Gamma variants. The female mounted robust and sustained anti-SARS-CoV-2 responses. Blood samples collected their after delivery verified prenatal high levels spike-specific IgG, which positively correlated IgG titers at term. In addition, an in vitro test ACE2 neutralization indicated that infants' demonstrated antigen specificity, reflecting immunization either Wuhan Gamma-variant antigens. All sera showed stronger ACE2-RBD binding inhibition when variants assay more closely resembled vaccine RBD sequence than distantly related (i.e., Delta Omicron). Monkeys are a valuable animal for evaluating new vaccines promote infant health. Further, findings highlight enduring nature safety elicited by recombinant RBD-Fc vaccine.
