Abstract
Endothelium,
the
lining
in
this
blood
vessel,
orchestrates
three
main
critical
functions
such
as
protecting
components,
modulating
of
hemostasis
by
secreting
various
inhibitors,
and
directing
clot
digestion
(fibrinolysis)
activating
tissue
plasminogen
activator.
No
other
surface
can
perform
these
tasks;
thus,
contact
blood‐contacting
medical
devices
inevitably
leads
to
activation
coagulation,
often
causing
device
failure,
thromboembolic
complications.
This
perspective,
first,
discusses
biological
mechanisms
coagulation
highlights
efforts
advanced
coatings
recapitulate
one
characteristic
endothelium,
hereafter
single
endothelium
noting
necessity
synergistic
integration
its
functions.
Subsequently,
it
is
emphasized
that
overcome
challenges
compatibility
an
endothelium‐mimicking
system
needed,
proposing
a
synergy
bottom‐up
synthetic
biology,
particularly
cells,
with
passive‐
bioactive
coatings.
Such
holds
promise
for
developing
biomaterials
capable
recapitulating
endothelial
functions,
thereby
enhancing
hemocompatibility
performance
devices.

Existing
anticoagulant
coatings
are
designed
to
inhibit
the
initial
attachment
of
coagulant
factors,
but
they
frequently
suffer
from
undesirable
coating
defects
that
result
in
a
declined

barrier
function.
Heparin,
serving
as
commercialized
pharmacological
via
activation
antithrombin,
causes
heparin-induced
thrombocytopenia.
Here,
we
developed
an
argatroban-polyphenol
conjugate
proteinaceous
(named
PDA-BSA-AG
coating)
actively
inhibits
thrombin,
contrast
previously
reported
passive
and
indirect
anticoagulants.
Following
clinical
treatment
standards,
magnetic
levitation
ventricular
assist
system
was
employed
for
first
time
accomplish
extracorporeal
circulation
utilizing
coated
tubes.
The
results
show
clotting
68.9
%
longer
than
Bioline
heparin
coating.
It
is
noteworthy
argatroban
not
only
delivers
effectiveness
also
enhances
patient
safety
reduces
occurrence
complications.
A
novel
combination
primary
specific
proteins

coagulation-inflammatory
cascade
reaction
cytokines
complement
systems
explained
anticoagulation
mechanism.
Reliable
biocompatibility
(hemolysis,
cytotoxicity,
antibacterial
activity)
noted,

impressive
