Liposarcoma is the most common soft tissue sarcoma, and surgical resection primary treatment. However, liposarcoma recurrence delayed wound healing remain critical challenges due to extensive interventions. Herein, we design an injectable polyethylene glycol (PEG)-based hydrogel (PEG@MT) with high skin adhesion loading melatonin, concurrently prevent promote postoperative healing. The formed by mixing 8-arm-polyethylene glycol-succinimidyl glutarate (8-arm-PEG-SG) 4-arm-polyethylene glycol-amine (4-arm-PEG-NH2) at a 1:1 ratio, establishing amide-bond crosslinked network. Excess succinimidyl ester groups bind endogenous amine groups, ensuring for effective closure. Functional assays demonstrated its dual functionality: promoting angiogenesis while suppressing proliferation metastasis. Mechanistically, PEG@MT promotes cell cycle arrest apoptosis via FOXO pathway in cells. studies also established novel model validate therapeutic outcomes. offers closure of capacity. Meanwhile, sustained release melatonin wounds significantly inhibits local liposarcoma. Overall, developed capable preventing shows valuable application postsurgical liposarcoma-resection management.
