Abstract Background Hepatocellular carcinoma (HCC) poses ongoing difficulties for public health systems due to its high incidence and poor prognosis. Huqi formula (HQF), a well-known prescription in traditional Chinese medicine, has demonstrated notable clinical effectiveness the treatment of HCC. However, mechanisms underlying therapeutic effects have yet be completely elucidated. Purpose This study aimed investigate anti-HCC HQF mechanisms. Methods Chemical profiling quantification were conducted by LC–MS HPLC. Orthotopic subcutaneous tumor models established through hydrodynamic injection Akt/Nras plasmids c-Met/sgPten cells, respectively, evaluate on Network pharmacology, RNA-Seq, molecular docking, Western blot, flow cytometry employed assess Results analysis identified 41 components, with HPLC showing salvianolic acid B as most abundant compound (0.303%). In c-Met/sgPten-induced HCC models, significantly reduced tissue damage, improved liver function, inhibited progression. Mechanistic studies revealed that induced apoptosis cells downregulating p-PI3K, p-AKT, p-mTOR expression, docking indicating strongest binding affinity between PI3K. further enhanced CD4 + CD8 T cell infiltration within microenvironment.
