Abstract Drug‐resistant microorganisms cause serious problems in human healthcare, leading to the persistence infections and poor treatment outcomes from conventional therapy. In this study, a gene‐targeting strategy using microbubble‐controlled nanoparticles is introduced that can effectively eliminate biofilms of methicillin‐resistant Staphylococcus aureus (MRSA) vivo. Biofilm‐targeting (BTN) capable delivering oligonucleotides are developed remove biofilm‐associated bacteria upon controlled delivery with diatom‐based microbubblers (MB). The activity BTN silencing key bacterial genes related MRSA biofilm formation ( icaA ), growth ftsZ antimicrobial resistance mecA as well their multi‐targeting ability vitro validated. integration MB next examined, resulting synergistic effects removal an ex vivo porcine skin model. therapeutic efficacy further investigated mouse wound model infected biofilm, which showed MB‐controlled substantially reduced load led effective elimination biofilm. This study underscores potential gene platform physical enhancement promising combat antibiotic resistance.
