DNA's ability to exist in a wide variety of structural forms, subforms, and secondary motifs is fundamental numerous biological processes has driven the development biotechnological applications. Major determinants DNA flexibility are multiple torsional degrees freedom around phosphodiester backbone. This high complexity can be rationalized by using two pseudotorsional angles linking atoms P C4′, from which Ramachandran-like plots built. In this contribution, we explore distribution η (eta: C4′i–1-Pi-C4′i-Pi+1) θ (theta: Pi-C4′i-Pi+1-C4′i+1) known experimental structures retrieved Protein Data Bank (PDB), subdividing conformational space into different datasets. Targeted delivery of chemotherapeutic agents is a well-established approach to cancer therapy. Antibody-drug conjugates (ADCs) typically carry toxic payloads attached tumor-associated antigen-targeting IgG antibody via an enzyme-cleavable linker that releases the drug inside cell. Aptamers are promising alternative antibodies in terms antigen targeting; however, their polynucleotide nature and smaller size result completely different PK/PD profile compared IgG. This may prove advantageous: owing lower molecular weight, aptamer-drug achieve better penetration solid tumors ADCs.
