Gene-editing technology has become a transformative tool for the precise manipulation of biological genomes and holds great significance in field animal disease-resistant breeding. Mastitis, prevalent disease husbandry, imposes substantial economic burden on global dairy industry. In this study, regulatory sequence gene editing breeding strategy successful creation gene-edited (GED) goats with enhanced mastitis resistance using ISDra2-TnpB system as model is proposed. This included targeted integration an innate inflammatory (IRS) into promoter region lysozyme (LYZ) gene. Upon Escherichia Coli (E. coli) mammary gland infection, GED exhibited increased LYZ expression, showing robust anti-mastitis capabilities, mitigating PANoptosis activation, alleviating blood-milk-barrier (BMB) damage. Notably, highly expressed only E. coli infection. study marks advent animals exogenous-free expression demonstrates feasibility gene-editing proposed study. addition, it provides novel blueprint developing strains, focusing specificity biosafety while providing research basis widespread application system.
