Abstract Inspired by the advantages of biological cascade catalytic systems, it has been devoted to discovery novel oxidase‐based systems for disease monitoring. However, low stability, easy inactivation, and poor reproducibility oxidase significantly limit their practical applications. Immobilization can be enabled protect them from external mediators improve efficiency reproducibility. Notably, substrate channels spatial confinement play an essential role in construction immobilized nanoreactors enhance overall activity. Moreover, nanozymes, a class enzyme mimics, have not only enzyme−like activity but also high stability tunable properties, which bolster development nanoreactors. Herein, recent advances assembly reactors involving enzymes/nanozymes are described. The importance channeling distribution regulating nanoreactor is highlighted. Then, along with in−depth discussion biosensors monitoring, design application innovative devices based on these sensing principles summarized, including microfluidic hydrogel−based platforms, test paper technologies. Finally, challenges prospects briefly discussed prospected.
