Abstract Malignant tumor has become one of the hellish killers threatening health people around world, its diagnosis and treatment concerns public. However, optimal therapeutic dose, undesired side‐effect, long‐term immune activation were key bottleneck problems in treatment. Herein, different batches supramolecular platforms, including vesicles, spherical nanoparticles, cylindrical nanorods, with precise ratios dye to drug (1:2) multiple stimulus responsiveness constructed by host‐guest complexation between cyanine‐camptothecin conjugates (IR780‐CPT2) β‐cyclodextrin (β‐CD) pendent hydrophilic copolymers. The reduction responsiveness, near‐infrared photothermal conversion singlet oxygen ( 1 O 2 ) generation performances endowed these platforms excellent cancer cells killing effect both vitro cellular experiments vivo mice models. More importantly, without affecting weight mice, maturation dendritic cells, proliferation T up‐regulation high mobility group protein B1, immunosuppressive regulatory detected after employing a synergistic chemo‐photo therapy, demonstrating body's was successfully activated. Thus, during primary tumor, distal also inhibited. We believe this work could provide distinctive way fabricate theranostic morphologies improve antitumor antimetastasis capabilities.

Constructing J-aggregated organic dyes represents a promising strategy for obtaining biomedical second near-infrared (NIR-II) emissive materials, as they exhibit red-shifted spectroscopic properties upon assembly into nanoparticles (NPs) in aqueous environments. However, currently available NIR-II J-aggregates primarily rely on specific molecular backbones with intricate design strategies and are susceptible to fluorescence quenching during assembly. A facile approach constructing bright using prevalent donor-acceptor (D-A) molecules is still lacking. In this study, we present method that transforms D-A by simply bending the molecule through introducing methyl group, enabling high-performance phototheranostics. The TAA-BT-CN exhibits hypsochromic-shift absorption forming H-aggregated NPs, while designed mTAA-BT-CN bent structure demonstrates bathochromic shift of over 100 nm leading much enhanced emission beyond 1100 nm. With respect its counterpart aggregation-caused (ACQ) phenomenon, NPs 7-fold increase owing their aggregation-induced (AIE) property well efficient generation heat reactive oxygen species under 808 light excitation. Finally, employed whole-body blood vessel imaging technology imaging-guided tumor phototherapies. This study will facilitate flourishing advancement based D-A-type molecules.
