Azobenzenes (Ph-N═N-Ph) are known as the most widely studied molecular photoswitches, and recent rise of azoheteroarenes (Het-N═N-Ph or Het-N═N-Het) offers great opportunities to advance this already mature field. A common limitation is that azo-switches generally require harmful UV light for activation, which hinders their application across various fields. Despite efforts in developing visible-light azobenzenes over past few decades, potential heteroaryl azoswitches remains largely unexplored. This Perspective summarizes state-of-the-art advancements azoswitches, covering design strategies, structure-property relationship, applications. We highlight distinctive advantages research development switches. Furthermore, we discuss challenges emerging field propose solutions address crucial issues such spectral red-shift thermal half-life. Abstract Molecular photoswitches can undergo isomerization under light exposure, making them uniquely attractive for high spatiotemporal resolution remote control of biological functions. Visible and near‐infrared (NIR) light, with their low energy consumption, safety, strong tissue penetration, are particularly suitable applications. Therefore, developing visible‐ NIR light‐driven molecular use is great significance. This review introduces the most common presents state‐of‐art applications in bioimaging, biosensing, drug delivery, photocontrolled cancer phototherapy, photopharmacology. The opportunities challenges future development outlined.

Non-alcoholic fatty liver disease (NAFLD) is the hepatic manifestation of metabolic syndrome. NAFLD encompasses a spectrum damage starting with steatosis and lipid disorders presented as hallmark. Cannabinoid-2 receptor (CB2R) endocannabinoids mainly expressed in immune cells. Our preliminary study revealed preventative role CB2R injury related to metabolism. In this study, we aimed explore underlying mechanism microbial community. High-fat diet-induced model was established mice. We found that expression significantly reduced mice CB2R-/- fed normal chow. Interestingly, cohousing or transplanted microbiota from WT mice, treatment an antibiotic cocktail ameliorated phenotype The gut dysbiosis including increased Actinobacteriota decreased Bacteroidota similar patients Microbial functional analysis metabolomics profiling obviously disturbed tryptophan metabolism which were also seen Correlation network showed disordered metabolites such indolelactic acid (ILA) xanthurenic mediated by severity indicators. vitro vivo validation experiments enriched ILA aggravated phenotypes. These results demonstrate involvement NAFLD, microbiota-mediated metabolites. findings highlight associated microbes promising targets for NAFLD.

Class A G protein‐coupled receptors (GPCRs) continue to garner interest for their essential roles in cell signalling and importance as drug targets. Although numerous drugs the clinic target these receptors, over 60% GPCRs remain unexploited. Moreover, adverse effects triggered by available unbiased GPCR modulators, limit use therapeutic value. In this context, elucidation of biased has opened up new pharmacological avenues holding promise safer therapeutics. Functionally selective ligands favour receptor conformations facilitating recruitment specific effectors modulation associated pathways. This review surveys current discovery landscape GPCR‐biased modulators with a focus on recent advances. Understanding biological preferential coupling is at different stages depending family.
