Abstract Photosensitizers with near‐infrared (NIR) excitation, especially above 800 nm which is highly desired for phototherapy, remain rare due to the fast nonradiative relaxation process induced by exciton‐vibration coupling. Here, a diketopyrrolopyrrole‐derived photosensitizer (DTPA‐S) developed via thionation of carbonyl groups within diketopyrrolopyrrole skeleton, results in large bathochromic shift 81 nm, endowing strong NIR absorption at 712 nm. DTPA‐S then introduced functional biomolecule (N 3 ‐PEG 2000 ‐RGD) click reaction construction integrin αvβ3 receptor‐targeted nano‐micelles (NanoDTPA‐S/RGD), endows further superlarge redshift 138 thus extending maxima ≈850 Remarkably, thiocarbonyl substitution increases nonbonding characters frontier molecular orbitals, can effectively suppress vibrational reducing reorganization energy, enabling efficient reactive oxygen species (ROS) generation under 880 excitation. Screened vitro and vivo assays, NanoDTPA‐S/RGD high water solubility, excellent tumor‐targeting ability, photodynamic/photothermal therapy synergistic effect exhibits satisfactory phototherapeutic performance. Abstract The reactive oxygen species (ROS) amplification caused by inevitable plasma albumin encapsulation is still a challenge to circumvent the systemic adverse effects in photodynamic therapy (PDT) process. Herein, disulfide bond linked homodimer, Cy1280, which modulated accurately balance fluorescence and ROS generation exhibit weak sealed PDT effect during blood circulation, exploited. Cy1280 can be specifically internalized dispersed at tumor site via Organic Anion Transporter Proteins (OATPs) thiol‐disulfide exchange mediated synergistic uptake activated after mild sunlight irradiation (100 ± 5 Klx) sensitize neighboring cellular mitochondria execute direct protein dysfunction effect. dynamic covalent chemistry (DCC) facilitates prolonged sustained retention tumors (>336 h) demonstrates efficacy of imaging‐guided solid‐tumor tumor‐bearing BALB/C mice. Most photosensitizers face enormous challenges in tumor hypoxia, the redox microenvironment, and low immune efficacies for reactive oxygen species (ROS). Herein, dye SQ-580 was constructed by coupling electron-donating indole thiophenazine-thiophene with electron-withdrawing dicyanovinyl squaraine. It exhibited a high generation of •OH O2•- decreasing ΔES1T2 acted as an excellent type I photosensitizer conquering hypoxia. The nanoplatform involving SQ-580, MnO2, targeting peptide CREKA targeted breast tumor. In MnO2 reacted high-expressed GSH produced Mn2+, which catalyzed H2O2 to decompose into induced chemodynamic therapy (CDT). reduction inhibited consumption maintained its photodynamic (PDT) efficacy. depletion ROS resulted cell ferroptosis. Under synergy ferroptosis ROS, Mn2+ amplified immunogenic death (ICD). mouse models, SQ-580@MnO2 NPs showed NIRF/MR imaging-guided targeting, effectively growth primary distant tumors, PDT PDT, CDT, ferroptosis, ICD. With the widespread application of tagging materials, existing chemical materials exhibit limitations in stability and detection under field conditions. This study introduces a novel magnetic scheme. Hydrophilic material-modified Fe3O4 nanoparticles (COOH-PEG@Fe3O4 NPs) were synthesized using co-precipitation technique.
