Abstract Polymethine cyanine dyes have witnessed significant advancements in bioimaging and phototherapy due to their optical properties ease of modification. Dysfunction of subcellular organelles initiates complex pathophysiological cascades and underlies numerous diseases, underscoring the need for organelle-specific therapeutic interventions. Precise spatiotemporal control reactive oxygen species (ROS) generation within offers a promising intervention approach. Herein, we report design synthesis novel series organelle-targeted, photoactivatable acetylperoxyl radical donors (ACR575s) based on an acetyl-caged rhodamine scaffold. Blue light irradiation triggered release highly oxidative radicals, concomitantly generating dye real-time monitoring. In vitro studies demonstrated delivery which subsequently induced concentration-dependent stress specific compartments. Notably, this resulted in membrane damage modulation macrophage polarization, providing clear evidence potential radicals regulating redox balance inflammatory responses. The ACR575 provides toolset biology regulation, enabling precise radical-mediated showing applications cancer therapy.
