Abstract Astragalus species are ubiquitous in the pharmacopeia of numerous countries, signifying their widespread medicinal applications. Türkiye is esteemed as one principal epicenters diversity for this genus with 483 taxa, and many these plants have been traditionally utilized diseases including coughing, diabetes, cardiovascular disorders, aches. Although there a considerable body chemical biological research available on several species, studies focusing brachystachys DC exceedingly rare. Attention-deficit/hyperactivity disorder (ADHD) is a persistent neurodevelopmental disorder. Despite pharmacological interventions, there need for effective lead molecules and therapeutic targets. Recently, Hericium erinaceus (HE) has been traditionally reported to treat various diseases. Herein, we aimed explore the noncytotoxic properties, phytochemical composition, spectroscopic characterization of HE aqueous extract. Additionally, used computational workflows identify key targets ADHD assess extract phytoconstituents potential targeting. Initially, was obtained using Soxhlet extraction, its cytotoxicity assessed on SH-SY5Y cells MTT assays. FTIR spectroscopy characterized extract's functional groups, while biochemical methods GC-MS identified constituents. A protein-protein interaction network targets, molecular docking, dynamics, QM/MM calculations were find drug candidates from As result, exhibited no in across concentrations (0.625 10 μg/mL) after 24 h. analysis detected 13 different groups that hold diverse biological importance. Qualitative screening revealed presence carbohydrates, flavonoids, anthocyanins, tannins, alkaloids, saponins, steroids, phenolic compounds. profiling 17 metabolites. Simultaneously, ADHD-related genes known protein integrated into network, identifying SLC6A4 as hub target. Molecular docking compounds showed myo-inositol's high binding efficiency (-6.53 kcal/mol). Dynamic simulations demonstrated stable interactions, confirmed ability transfer electrons, reinforcing potential. Overall, shows potent nontoxic profile contains like myo-inositol, offering promising by targeting ADHD.
