Abstract Loz1 is a zinc‐responsive transcription factor in fission yeast that maintains cellular zinc homeostasis by repressing the expression of genes required for uptake high conditions. Previous deletion analysis found region containing two tandem C 2 H zinc‐fingers and an upstream “accessory domain” rich histidine, lysine, arginine residues to be sufficient zinc‐dependent DNA binding gene repression. Here we report unexpected biophysical properties this pair seemingly classical fingers. Isothermal titration calorimetry NMR spectroscopy reveal distinct events localized spectra complex dynamic behavior spanning time scales from fast 10 −12 –10 −10 slow >10 0 s. Slow exchange due cis‐trans isomerization TGERP linker results doubling many signals protein. Conformational on −3 s timescale throughout first finger distinguishes it second linked weaker affinity zinc. These findings mechanism sensing illuminate how protein's rough free‐energy landscape enables sensing, regulated expression.

BTB (bric-a-brack, Tramtrack, and broad complex) is a diverse group of protein-protein interaction domains found within metazoan proteins. Transcription factors contain dimerizing subtype with characteristic N-terminal extension. The Tramtrack (TTK) distinct type domain, which can multimerize. Single-particle cryo-EM microscopy revealed that the TTK-type assemble into hexameric structure consisting three canonical dimers connected through previously uncharacterized interface. Zanthoxylum armatum (Z. armatum) is a significant economic tree species known for its medicinal and edible properties. However, the presence of prickles on Z. poses considerable challenge to advancement industry. Numerous studies have indicated that C2H2 zinc finger protein (C2H2-ZFPs) families are crucial in development plant trichomes or prickles. This study identified 78 ZaC2H2 genes from genome, categorizing them into three groups analyzing their physicochemical properties, chromosomal locations, conserved domains, gene structures. The evolutionary analysis indicates amplification primarily results whole-genome duplication segmental duplication, these undergone strong purifying selection pressure throughout history. cis-acting elements revealed they contain various hormone response elements, such as ABRE, AuxRR, CGTCA motif, GARE motifs, TCA which responsive ABA, IAA, MeJA, GA, SA signals. RT-qPCR was employed assess expression levels candidate ZaC2H2-45, ZaC2H2-46, ZaC2H2-49, ZaC2H2-55 under treatment five hormones. ZaC2H2-46 were significantly up-regulated NAA, SA, MeJA treatments. These will help further understand characteristics family provide theoretical basis studying
