Alzheimer’s disease (AD) and Parkinson’s (PD) are two typical neurodegenerative diseases that increased with aging. With the emergence of aging population, health problem economic burden caused by also increase. Phosphatidylinositol 3-kinases/protein kinase B (PI3K/AKT) signaling pathway regulates signal transduction biological processes such as cell proliferation, apoptosis metabolism. According to reports, it neurotoxicity mediates survival neurons through different substrates forkhead box protein Os (FoxOs), glycogen synthase kinase-3β (GSK-3β), caspase-9. Accumulating evidences indicate some natural products can play a neuroprotective role activating PI3K/AKT pathway, providing an effective resource for discovery potential therapeutic drugs. This article reviews relationship between AKT AD PD, discusses based on treat in recent years, hoping provide guidance reference this field. Further development Chinese herbal medicine is needed these diseases.

Doxorubicin (DOX) is a well-known chemotherapeutic agent extensively applied in the field of cancer therapy. However, similar to other agents such as cisplatin, paclitaxel, docetaxel, etoposide and oxaliplatin, cells are able obtain chemoresistance that limits DOX efficacy. In respect dose-dependent side effect DOX, enhancing its dosage not recommended for effective chemotherapy. Therefore, different strategies have been considered reversing resistance diminishing effects. Phytochemical potential candidates this case due their great pharmacological activities. Curcumin antitumor phytochemical isolated from Curcuma longa with capacity suppressing metastasis proliferation affecting molecular pathways. Experiments demonstrated curcumin inhibiting by downregulating oncogene pathways MMP-2, TGF-β, EMT, PI3K/Akt, NF-κB AP-1. Furthermore, coadministration potentiates apoptosis induction cells. light this, nanoplatforms employed codelivery DOX. This results promoting bioavailability internalization aforementioned active compounds and, consequently, activity. Noteworthy, has reducing adverse effects on normal tissues via inflammation, oxidative stress apoptosis. The current review highlights anticancer mechanism, nanovehicles.

The increase in human life expectancy has become a challenge to reduce the deleterious consequences of aging. Nowadays, an increasing number population suffer from age-associated neurodegenerative diseases including Parkinson's disease (PD) and Alzheimer's (AD). These disorders present different signs neurodegeneration such as mitochondrial dysfunction, inflammation, oxidative stress. Accumulative evidence suggests that transcriptional factor nuclear (erythroid-derived 2)-like 2 (Nrf2) plays vital defensive role orchestrating antioxidant response brain. Nrf2 activation promotes expression several enzymes exert cytoprotective effects against damage impairment. In this context, studies have proposed pathogenesis PD AD. Thus, we consider it important summarize ongoing literature related pathway context these diseases. Therefore, review, discuss mechanisms involved activity its connection with mitochondria, energy supply, Furthermore, will lead our discussion identify participation impairment Finally, therapeutic could on cognitive impairment, neurodegeneration, failure

This review discusses the impact of curcumin—an aromatic phytoextract from turmeric (Curcuma longa) rhizome—as an effective therapeutic agent. Despite all beneficial health properties ensured by curcumin application, its pharmacological efficacy is compromised in vivo due to poor aqueous solubility, high metabolism, and rapid excretion that may result systemic bioavailability. To overcome these problems, novel nanosystems have been proposed enhance bioavailability bioactivity reducing particle size, modification surfaces, encapsulation efficiency with different nanocarriers. The solutions based on nanotechnology can improve perspective for medical patients serious illnesses. In this review, we discuss commonly used curcumin-loaded bio-based nanoparticles should be implemented overcoming innate constraints natural ingredient. Furthermore, associated challenges regarding potential applications combination therapies are discussed as well.
