In recent years, there has been a global increase in the prevalence of allergic diseases, including rhinitis, chronic rhinosinusitis, asthma, atopic dermatitis, conjunctivitis, and food allergies. Since pathogenic mechanisms these diseases are not yet fully understood, targeted effective therapies lacking. The NLRP3 inflammasome, multiprotein complex implicated various inflammatory can be activated by diverse stimuli. It assembles into inflammasome complexes through conformational changes, initiating proteolytic cleavage dormant procaspase-1 active caspase-1 promoting maturation cytokines, IL-1β IL-18. Dysfunction may serve as key driver leading to pyroptosis amplifying local response. Abstract Background and aim Ulcerative colitis (UC) is characterized by complex immunological interactions involving CD4 T cell subsets the NLRP3 inflammasome, which influence inflammatory responses. This investigation focused on delineating activation profiles of these components their correlation with disease severity activity, assessing diagnostic implications in UC. Methods We conducted immunohistochemistry ELISA assays to measure markers expression inflammasome UC patients versus controls. Findings were validated using analysis, molecular docking ROC curves. Results displayed increased Th1 (T-bet, TNF-α), Th2 (GATA3, IL-6), Th17 (RORγt, IL-17, IL-22, IL-23) Additionally, cytokines (IL-2 IL-4) significantly elevated severe UC, while Treg (FOXP3, IL-10, TGF-β1) only mild-to-moderate Enhanced activation, indicated NLRP3, Caspase-1, IL-1β levels. These patterns, confirmed through analysis docking, underscored strong correlations among T-bet, GATA3, supporting proposed NLRP3/T-bet/GATA3 axis. axis, along other biomarkers, showed associations severity, Mayo score, UCEIS, demonstrated relatively high value. Conclusion The axis provides a referable strategy for multi-targeted combined treatment may serve as potential biomarkers enhancing accuracy guiding therapy.

Allergic rhinitis (AR) is a chronic, non-infectious inflammatory condition of the nasal mucosa mediated by IgE. There need for development novel medications to treat this ailment. Isoorientin naturally occurring flavonoid that possesses antioxidant, anti--inflammatory, and various other advantageous characteristics. However, its potential effects on AR remain unclear. This study evaluates therapeutic isoorientin ovalbumin (OVA)-induced allergic in mice explores underlying mechanism. Our revealed administration effectively decreased frequency nose rubbing sneezing mice. The groups treated with showed significant decrease serum levels IgE histamine, reductions 40% 30%, respectively. ameliorated inflammation restored Th1/Th2 balance. In addition, inhibited activation NF-κB pathway tissues. summary, alleviates OVA-stimulated restoring balance blocking pathway. Thus, exhibits promise as natural agent rhinitis.
