The recent relaxation of psychedelic drug regulations has prompted extensive clinical investigation into their potential use to treat diverse mental health conditions including anxiety, depression, post-traumatic stress, and substance-abuse disorders. Most trials have relied on a small number known molecules found in nature, such as psilocybin, or long-known synthetic analogs natural metabolites, lysergic acid diethylamide (LSD). Elucidation biosynthetic pathways leading several compounds established an opportunity biology complement chemistry for the preparation novel derivatives with potentially superior pharmacological properties compared drugs. Herein we review metabolic biochemistry from plants, fungi animals that yield medicinally important hallucinogenic specialized metabolites ibogaine, mescaline, acid, N,N-dimethyltryptamine (DMT). We also summarize reconstitution these microorganisms comment integration native non-native enzymes prepare derivatives.
