Abstract Alterations in the experience-dependent and autonomous elaboration of neural circuits are assumed to underlie autism spectrum disorder (ASD), though it is unclear what synaptic traits responsible. Here, utilizing a valproic acid–induced ASD marmoset model, which shares common molecular features with idiopathic ASD, we investigate changes structural dynamics tuft dendrites upper-layer pyramidal neurons adjacent axons dorsomedial prefrontal cortex through two-photon microscopy. In model marmosets, dendritic spine turnover upregulated, spines generated clusters survived more often than control marmosets. Presynaptic boutons local axons, but not commissural long-range demonstrate hyperdynamic suggesting alterations projection-specific plasticity. Intriguingly, nasal oxytocin administration attenuates clustered emergence Enhanced generation, possibly unique certain presynaptic partners, may be associated potential therapeutic target.

Abstract Oxytocin (OXT) is a peptide hormone and neuropeptide that regulates various peripheral physiological processes modulates behavioral responses in the central nervous system. While humoral release occurs from axons arriving at median eminence, also released oxytocinergic cell brain structures contain its receptor, their dendrites hypothalamic nuclei potentially into cerebrospinal fluid (CSF). Understanding oxytocin’s complex functions requires knowledge on patterns of projections relationship to receptor (OXTR). This study provides first comprehensive examination system prairie vole ( Microtus ochrogaster ), an animal exhibiting social behaviors mirror human linked functioning. Using light electron microscopy, we characterized neuroanatomy this species. OXT+ bodies were found primarily hypothalamus, densest subcortical regions. Examination fibers oxytocin transcripts Oxtr ) revealed except for some structures, presence was not correlated with amount across brain. Of particular interest, cerebral cortex had high expression contained little no fibers. Electron microscopy used quantify dense cored vesicles (DCV) identify potential axonal sites. The ependymal cells line ventricles frequently permissive DCV-containing reaching third ventricle. Our results highlight mechanism which directly circulates throughout ventricular system, may serve as primary source binds OXTR cortex.

Early life sleep is important for neuronal development. Using the highly social prairie vole rodent model, we have previously reported that early disruption (ELSD) during preweaning period results in interference with bonding and increases ethanol consumption following a stressor adulthood. Furthermore, ELSD parvalbumin expression reduces glutamatergic neurotransmission cortical regions adult voles. To understand impact of on lifespan, an examination earlier time necessary. The aim present study was to examine behavioral outcomes adolescent Given known effects development systems involved mood motivation, hypothesized anxiety, risk, reward-related behaviors would be impacted by We report both male female voles experienced showed heightened anxiety-like behavior compared age-matched controls (CONs) as measured light-dark box. Additionally, reductions preference consumption, affiliative CONs. These suggest sexes experience reduced reward-seeking after ELSD. further critically neurotypical adolescence.
