The article aimed at discussing an alternative option, non-invasive vagus nerve stimulation (VNS), roles it plays pathology immunotherapy. VNS does this reducing oxidative stress via silent information regulator 1 (SIRT1); inhibiting inflammation both hypothalamic-pituitary-axis (HPA) release corticosteroid from adrenal gland, cholinergic anti-inflammatory pathway (CAP), increasing vagal activity which helps regulation cell proliferation, differentiation, apoptosis, metabolism, increase chance survival. To treat diseases associated with vagal nerve control of peripheral organs, it is necessary to selectively activate efferent and afferent fibers in the vagus. Activation of the vagus nerve mediates cardioprotection and attenuates myocardial ischemia/reperfusion (I/R) injury. In response to vagal activation, acetylcholine (ACh) is released from intracardiac nervous system (ICNS) activates intracellular cardioprotective signaling cascades. Recently, however, a nonneuronal cholinergic cardiac (NNCCS) in cardiomyocytes has been described as an additional source ACh. To investigate whether NNCCS absence ICNS we used reductionist approach isolated adult rat ventricular without neuronal cells, using hypoxic preconditioning (HPC) protective stimulus. Adult were isolated, cells was confirmed, HPC induced by 10/20 min hypoxia/reoxygenation (H/R) before subjection 30/5 H/R simulate I/R Cardiomyocyte viability assessed trypan blue staining at baseline after HPC+H/R or H/R. Intra- extracellular ACh quantified liquid chromatography-coupled mass spectrometry baseline, HPC, hypoxia, reoxygenation, respectively. subset experiments, muscarinic nicotinic receptor (m- nAChR) antagonists added during (69 ± 4%) reduced (10 3%). With cardiomyocyte preserved (25 6%). increased hypoxia; further both intra- (from 0.9 0.7 1.5 1.0 nmol/mg; 0.6 1.1 nmol/mg, respectively).
