Solid-state synthesis is an approach to organic that desirable because it can offer minimal or no solvent waste, high yields, and relatively low energy footprints. Herein, we report the solid-state of a novel Schiff base, 4-{(E)-[(4-methylpyridin-3-yl)imino]methyl}benzoic acid (4-PIBZ), synthesized through reaction amine aldehyde. 4-PIBZ was prepared via solvent-drop (water) grinding (SDG) on multigram scale with 97% yield characterized using FTIR, 1H NMR, SCXRD. The pyridyl carboxylate moiety present in make suitable for use as linker ligand indeed found coordinate Cu(II), Zn(II), Cd(II) cations, enabling serve assembly coordination polymers. thereby formed 1D (spiro chain) 2D (square lattice, sql, topology) polymers solvent-induced (layering slurry) methods. resulting were though X-ray diffraction (SCXRD, PXRD) TGA, further demonstrating utility green methods preparation some classes new ligands turn be used

Solid-phase peptide synthesis (SPPS) is a ubiquitous methodology for making peptides from R&D to manufacturing plant, not least in the context of using and peptidometics as pharmaceuticals. Nevertheless, SPPS has long been plagued by its reliance on large amounts hazardous materials such dimethylformamide solvent trifluoroacetic acid cleavage reagent. Consequently, with methods producing sustainable manner being high demand, numerous reports covering various aspects greening have appeared recent years. In this chapter, an integrated overview state-of-the-art field presented, highlighting advances within entire workflow starting amino coupling cycle cleaving off polymer support. This work reports the first solvent-free catalytic approach for cleavage of fluorenylmethoxycarbonyl (Fmoc) protecting group from amine and alcohol functionalities. Various saccharide, peptide, glyco-amino acid substrates were efficiently deprotected by simple treatment with 20 mol % neat 4-dimethylaminopyridine (DMAP) (one effective base catalysts found), without any solvent or stoichiometric additives. Small model structures finally assembled through one-pot, base-catalyzed, multistep sequences combining Fmoc esterification, amidation, and/or glycosylation steps.

The widely used Fmoc/t-Bu solid-phase peptide synthesis (SPPS) is hampered by relying on corrosive, per/polyfluoroalkyl substance (PFAS) classified trifluoroacetic acid (TFA) as a universal protecting group (PG) removal/resin cleavage reagent. We report that suitable combinations of Brønsted acids (BAs) and Lewis (LAs) such HCl/FeCl3 AcOH/FeCl3 constitute viable alternatives for TFA PFAS-free agents. Using water miscible dimethyl carbonate (DMC) acetonitrile (MeCN) solvents enabled diluting mixtures with aqueous solutions, allowing direct use in purification which removal >99.99% iron from an induced was demonstrated.

Peptide therapeutics have exploded in popularity recent years, motivating the need for advanced manufacturing methods which can be applied across solid-phase peptide synthesis (SPPS) process. Heterobimetallic Pt(II)/Cu(II) single-chain polymer nanoparticles (SCNPs) were sequentially synthesized from a polymeric precursor featuring both α-diazo-β-ketoester and naked β-ketoester functional groups. Photoactivated carbene generation at λexc = 365 nm moieities was triggered for bonding Pt(II) ions dichloro(1,5-cyclooctadiene)Pt(II) to the precursor, whereas Cu(II) subsequently incorporated via Cu(II)-(β-ketoester)2 complex formation using acetate. Both intrachain complexation found contribute folding of generating Pt(II)/Cu(II)-SCNPs as evidenced by infrared spectroscopy, size exclusion chromatography dynamic light scattering. These heterobimetallic SCNPs proved highly efficient soft nanocatalysts consecutive one-pot alkyne semihydrogenation/alkene dioxygenation reactions room temperature in N-butylpyrrolidone, non-toxic alternative solvent N,N-dimethylformamide.
