eDNA metabarcoding has been used for the biomonitoring of benthic invertebrates, but correct steps to achieve its effectiveness, stability results, and comparisons with morphological methods are still understudied. In this study, morphology were studied invertebrate samples collected at six sites in Ulungu River Basin. A Mantel test NMDS analyses correlations communities obtained via two methods, Shannon index was calculated evaluate ecological status, correlation evaluation results analyzed. The revealed that did not detect a greater number species than method, from shared fewer taxa family level below, only five genus level. very significant differences between monitored by (PERMANOVA, p = 0.0056), provided richness abundance data difficult identify morphologically, these findings can be supplement data. cor.test there no diversity assessment cannot represent accurate true conditions. water sample eDNA-based method exhibited low consistency high complementarity monitoring diversity. More research is needed on key links sampling, control degradation rate, utilization, development provide more environmentally friendly effective protection, reliable support decision-making, adequately respond challenges global environmental change.
