Cadmium (Cd) can adversely damage plant growth. Therefore, understanding the control molecular mechanisms of Cd accumulation will benefit development strategies to reduce in plants. This study performed transcriptomic and metabolomic analyses on roots a Cd-tolerant Tartary buckwheat cultivar following 0 h (CK), 6 (T1), 48 (T2) treatment. The fresh weight root length were not significantly inhibited under T1 treatment but they T2 root's ultrastructure was seriously damaged evidenced by deformed cell walls, altered shape number organelles. A total 449, 999 differentially expressed genes (DEGs) eight, 37 metabolites (DEMs) identified CK versus comparison, respectively. DEGs analysis found that expression related wall function, glutathione (GSH) metabolism, phenylpropanoid biosynthesis changed during stress. ABSTRACT Female fertility has been demonstrated to be directly correlated with cadmium, a heavy metal contaminant that is widely present in the environment. N ‐acetyl cysteine (NAC), powerful antioxidant, shown have therapeutic effects for number of ailments. NAC's potential prevent ovarian toxicity caused by Cd still unknown. Therefore, objective current study was evaluate concentration‐ and time‐dependent protective effect NAC supplementation against Cd‐induced granulosa cell healthy caprine ovaries. The vitro cultured ovaries/follicles were subjected different cytotoxic (ethidium bromide acridine orange [EB/AO] staining), histomorphological, biochemical analyses during study. results revealed treatment significantly attenuated Cd‐instigated cytotoxicity cells, as evidenced diminished apoptotic attributes co‐supplemented groups compared only Cd‐exposed groups. Moreover, markedly restored decline enzymatic activity antioxidant enzymes (GST, SOD, CAT), along ferric reducing power, further diminishing formation MDA exposed ovary. findings this indicated Cd, being an toxicant, adversely affects female reproductive system. However, simultaneous administration reduced Cd‐caused damage, indicating controlling gonadotoxicity due gradually growing environmental pollution.

Graphene oxide (GO) has promising biomedical applications, but its potential toxicity to the female reproductive system is underexplored. This study investigates short-term effects of a single dose GO nanosheets on human ovarian granulosa cells, focusing mitochondrial damage. First, cell viability was detected by CCK-8 and apoptosis flow cytometry assess cytotoxicity KGN. Second, reactive oxygen species (ROS), membrane (MMP), morphology were observed confocal microscopy, sub-mitochondrial structure transmission electron microscopy (TEM), quantitative analysis ATP complex I enzyme activity luminosity value autophagy KGN cells. The 72h half-maximum effective concentration (EC50) determined be 29.73 μg/mL. induced death in dose-dependent manner, with significant even at low doses (1 μg/mL).
