Gastric cancer (GC) is a frequently diagnosed malignant tumor in clinical settings; however, the mechanisms underlying its tumorigenesis remain inadequately understood. In this study, we identified significantly elevated expression levels of AKR1B1 GC tissues through quantitative polymerase chain reaction (qPCR) and western blotting assays. Furthermore, negative correlation was established between patient survival probability levels. Functionally, our experiments, including colony formation, transwell migration, xenograft assays, demonstrated that depletion inhibited proliferation progression cells both vivo vitro. Additionally, assessment reactive oxygen species (ROS), malondialdehyde (MDA), glutathione (GSH), mitochondrial morphology confirmed induces ferroptosis. Mechanistically, found interacts with STAT3, which subsequently activates SLC7A11. Notably, ferroptosis induced by could be reversed overexpression SLC7A11, thereby substantiating these interactions. conclusion, findings identify as novel oncogene elucidate mechanism involving AKR1B1-STAT3-SLC7A11 pathway ferroptosis, providing new insights for potential therapeutic strategies treatment GC.

Ubiquitin-conjugating enzyme E2 T (UBE2T) is a crucial in the ubiquitin-proteasome system (UPS), playing significant role ubiquitination of proteins and influencing wide range cellular processes, including proliferation, differentiation, apoptosis, invasion, metabolism. Its overexpression has been implicated various malignancies, such as lung adenocarcinoma, gastric cancer, pancreatic liver ovarian where it correlates strongly with disease progression. UBE2T facilitates tumorigenesis malignant behaviors by mediating essential functions DNA repair, cell cycle regulation, activation oncogenic signaling pathways. High levels expression are associated poor survival outcomes, highlighting its potential molecular biomarker for cancer prognosis. Increasing evidence suggests that acts an oncogene could serve promising therapeutic target treatment. This review aims to provide detailed overview UBE2T’s structure, functions, mechanisms involved progression well recent developments UBE2T-targeted inhibitors.
