Abstract The tumour microenvironment (TME) constitutes the area surrounding during its development and has been demonstrated to play roles in cancer-related diseases through crosstalk with cells. Circular RNAs (circRNAs) are a subpopulation of endogenous noncoding (ncRNAs) that ubiquitously expressed eukaryotes have multiple biological functions regulation cancer onset progression. An increasing number studies shown circRNAs participate multifaceted TME. However, details on mechanisms involved remained elusive until now. In this review, we analyse effects TME from various perspectives, including immune surveillance, angiogenesis, hypoxia, matrix remodelling, exo-circRNAs chemoradiation resistance. Currently, enormous potential for circRNA use targeted therapy as noninvasive biomarkers drawn our attention. We emphasize prospect targeting an essential strategy regulate TME, overcome resistance improve therapeutic outcomes.

Background: Exosomal circular RNAs (circRNAs) in peripheral blood are considered as emerging diagnostic biomarkers of cancers. Circular RNAs are a large group of whose cellular functions still being investigated. We recently proposed that circSMARCA5 acts as sponge for the splicing factor Serine and Arginine Rich Splicing Factor 1 (SRSF1) in glioblastoma multiforme (GBM). After demonstrating by RNA immunoprecipitation physical interaction between SRFS1 circSMARCA5, we assayed real-time PCR cohort 31 GBM biopsies 20 unaffected brain parenchyma controls (UC) expression total, pro-angiogenic (Iso8a) anti-angiogenic (Iso8b) mRNA isoforms Vascular Endothelial Growth A (VEGFA), known target SRSF1. The Iso8a to Iso8b ratio: (i) increased with respect UC (p-value < 0.00001); (ii) negatively correlated (r-value = −0.46, p-value 0.006); (iii) decreased U87-MG overexpressing negative control 0.0055). Circular RNAs (circRNAs) are stable, often highly expressed RNA transcripts with potential to modulate other regulatory RNAs. A few circRNAs have been shown bind RNA-binding proteins (RBPs); however, little is known about the prevalence and distribution of these interactions in different biological contexts.We conduct an extensive screen circRNA-RBP ENCODE cell lines HepG2 K562. Abstract Background More and more studies have shown that circular RNAs (circRNAs) play a critical regulatory role in many cancers. However, the potential molecular mechanism of circRNAs prostate cancer (PCa) remains largely unknown. Methods Differentially expressed were identified by RNA sequencing. The expression hsa_circ_0003258 was evaluated using quantitative real-time PCR situ hybridization. impacts on metastasis PCa cells investigated series vitro vivo assays. Hsa_circ_0003258 may act promising biomarker for an attractive target intervention.

Previously, we have shown that apoplastic wash fluid (AWF) purified from Arabidopsis leaves contains small RNAs (sRNAs). To investigate whether these sRNAs are encapsulated inside extracellular vesicles (EVs), treated EVs isolated with the protease trypsin and RNase A, which should degrade located outside but not those inside. These analyses revealed mostly associated proteins. Further of (exRNAs) they include both long noncoding (lncRNAs), including circular (circRNAs). We also found exRNAs highly enriched in posttranscriptional modification N6-methyladenine (m6A). Consistent this, identified a putative m6A-binding protein AWF, GLYCINE-RICH RNA-BINDING PROTEIN 7 (GRP7), as well sRNA-binding ARGONAUTE2 (AGO2). two proteins coimmunoprecipitated lncRNAs, circRNAs. Mutation GRP7 or AGO2 caused changes sRNA lncRNA content suggesting contribute to secretion and/or stabilization exRNAs. propose mediate host-induced gene silencing, rather than RNA EVs.
