Abstract Evidence from preclinical animal models suggests that the stress-buffering function of endocannabinoid (eCB) system may help protect against stress-related reductions in hippocampal volume, as is documented major depressive disorder (MDD). However, stress exposure also lead to dysregulation this system. Thus, pathways marked histories, such childhood maltreatment (CM), smaller volumes and MDD humans depend on dysregulated versus intact eCB functioning. We examined whether relation between peripheral concentrations would vary a CM history. Further, we eCBs moderate CM/MDD volume. Ninety-one adults with 62 healthy comparison participants (HCs) were recruited for study Canadian Biomarker Integration Network Depression program (CAN-BIND-04). The eCBs, anandamide (AEA) 2-arachidonylglycerol (2-AG), assessed blood plasma. Severe history was retrospectively via contextual interview. associated though not all associations moderated by or direction expected. Specifically, higher AEA compared HCs regardless history, difference could be attributed psychotropic medications. 2-AG, but only CM. Consistent hypotheses, found lower left volume without CM, those AEA, high AEA. Our presents first evidence implicating mechanisms involving reduced MDD.

ABSTRACT Cannabidiol (CBD) and Δ 9 ‐tetrahydrocannabinol (THC), the main components of Cannabis sativa plants, can interact with specific cell receptors known as cannabinoid (CBs). The endogenous compounds anandamide (AEA) 2‐arachidonoylglycerol (2‐AG) are CB agonists, and, alongside enzymes, they constitute endocannabinoid system (ECS) take part in neuromodulation. Several LC‐MS/MS methods have been developed to quantify these biological matrixes, but a fast simple method that determine analytes plasma samples simultaneously is not available. Here, we propose disposable pipette extraction technique containing zirconia‐based sorbent (DPX(Zr)) combined UHPLC‐MS/MS analysis CBD, THC, AEA, 2‐AG samples, simultaneously. combines protein precipitation (PPT) one‐step DPX procedure remove phospholipids, one most common interferents samples. Optimization PPT‐DPX sample preparation reduced matrix effect improved sensitivity analytical method. validated DPX(Zr)‐UHPLC‐MS/MS reported LLOQs 0.1 ng mL −1 for AEA 1 CBD THC. demonstrated intra‐ interassay accuracy precision less than 20% LLOQ, 15% other calibration points. Additionally, no carryover or significant was observed. We applied this 2‐AG, obtained from obsessive‐compulsive disorder patients treated CBD.

The quantification of endocannabinoids in biological fluids is becoming increasingly popular as an indicator psychological and physiological function. Numerous methods to quantify the endocannabinoid ligands have been published so far, yet their concentrations responses often exhibit significant variability across studies. Endocannabinoids regulate interact with a wide range biomolecules, causing vary between cohorts individuals, sensitivities them depend on pre-experimental behaviours activities. Moreover, matrix effects produced by complex nature biofluids necessitate rigorous sample preparation techniques, all which introduce opportunities for both inter- intra-assay variability. This review aims address causes prior mass spectrometric analysis, including biofluid choice, human variability, collection extraction methods. If these factors are fully considered standardised introduced, may become more reliable, allowing utility clinical markers progress.

Anandamide (AEA) and 2-arachidonoylglycerol are endogenous agonists of the cannabinoid receptors regulate control many cellular functions. Their activities governed by enzymes proteins that their synthesis, receptor binding, transport, degradation, which known as endocannabinoid system (ECS). The aim this study was to investigate regulation activity in endometrium studying RNA protein expression ECS within endometrial cell types during different menstrual cycle stages impact endometriosis.
