Taraxacum officinale, a traditional medicinal herb, has garnered significant attention for its potential role in the prevention and treatment of breast cancer. Although clinical recognition efficacy gradually increased, research shown that officinale contains variety chemical components, including triterpenes, carbohydrates, flavonoids, phenolic acids, sesquiterpenes, coumarins, fatty organic acids. However, pharmacological mechanisms underlying officinale's effects identification key bioactive components warrant further investigation. Flow cytometry was utilized to investigate extract (TOE) combination with PD-1/PD-L1 inhibitor 2 on immune microenvironment triple-negative cancer (TNBC). Active compounds their targets were identified through an integrative approach involving GeneCards, OMIM, DisGeNET databases, as well UPLC-Q-Orbitrap MS analysis. Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) pathway enrichment analyses conducted, followed by molecular docking explore compound-target interactions. The anti-proliferative isochlorogenic acid A (ICGA-A) chicoric (CRA) MDA-MB-231 4T1 cells evaluated using CCK-8 assay. In vivo validation performed murine model flow cytometry. TOE active constituents, ICGA-A CRA, demonstrate augmenting PD-1 blockade therapy TNBC. This study investigated 2, which significantly enhanced infiltration macrophages CD8+ T into tumors models, while concurrently reducing population exhausted cells. Furthermore, CRA notably increased frequency Both therapies also found suppress tumor proliferation inhibiting FAK/PI3K/AKT/mTOR signaling pathway. These findings highlight effective adjuvants improve therapeutic inhibitor-based immunotherapy from exhibit antitumor activity TNBC targeting pathway, critical regulator progression. Their ability modulate highlights modulators enhance immunotherapy. suggest could serve promising adjuncts treatment, offering novel strategy overcome challenges such resistance limited options. Further investigation is warranted synergistic immunotherapies improving outcomes.

Despite the long history of Traditional Chinese Medicine (TCM) in disease treatment, underlying "Jun–Chen–Zuo–Shi" principle remains largely unexplored. To address this gap, it is essential to elucidate interactions between active substances TCM through quantitative molecular and cellular pharmacology. The Chou–Talalay method particularly effective for investigating drug combinations, making highly relevant formulas. This study employed explore drug–drug Xuebijing (XBJ), a formula used treating sepsis. aim was by main XBJ: danshensu salvianolic acid B (from Radix Salviae Miltiorrhizae), senkyunolide A Rhizoma Chuanxiong), ligustilide Angelicae Sinensis), safflower yellow hydroxysafflor Flos Carthami), paeoniflorin Paeoniae Rubra). We quantitatively analyzed their TLR4 antagonistic activities combination index (CI) quantify interactions, revealing synergism (CI < 1), additive effects = antagonism > 1). results show these agents inhibit nitric oxide (NO) production, with some combinations demonstrating synergistic at certain concentrations, while others present effects. Understanding provides scientific foundation optimizing formulations, enhancing quality control, efficacy, safety.

Artemisia annua L. (A. annua), a Traditional Chinese Medicine (TCM) that has been utilized in China for centuries, is known its potential anticancer properties. However, the main components and mechanism of action A. on endometrial carcinoma have not reported. We used TCMSP database to identify active their corresponding gene targets. then obtained targets specific cancer from The Cancer Genome Atlas (TCGA) GeneCards databases. common three databases were selected, "component-target" network was constructed. Protein–protein interaction (PPI) analysis ranking target proteins identified key protein PTGS2 analysis, PTGS2. also screened found quercetin, kaempferol, luteolin, isorhamnetin, artemisin, stigmasterol had most Molecular docking models established these six with PTGS2, revealing strong binding activity all them. Finally, we conducted validation experiments assess effects an component annua, cells (HEC-1-A Ishikawa cells). Our findings demonstrate quercetin inhibit both cell growth migration, while suppressing expression
