Abstract Human milk (HM) microbiota is increasingly studied for its potential health benefits. However, the physiological state of HM bacteria and consequently their effects on gut homeostasis remain a question. This study investigated by characterizing cultivable fraction, as it might be at point ingestion assessing digestion cultivability immunomodulatory properties six prevalent strains. The 28 samples was analysed 16S metabarcoding either directly raw (raw microbiota, RM) or complete fraction obtained from seven non-selective media (cultivable CM). Diversity lower in CM than RM, with 32 versus 435 genera median 7 69 per sample respectively. also showed under-representation strictly anaerobic genera. Factors like parity iron vitamin supplementation affected RM and/or CM. In vitro gastrointestinal moderately impacted strain cultivability. most strains partially completely lost monocyte THP1 cell line after digestion, except Staphylococcus epidermidis that gained potential.

The commensal microbiota influences the health, feeding efficiency, and reproductive performance of sows. composition in alimentary genitourinary tracts colostrum/milk during pregnancy lactation also impacts immune system, growth, health piglets. Knowledge microbial compositions is important for evaluation these effects discovering ways to improve productivity Oral, vaginal, rectal mucosa colostrum were sampled from 32 sows variable parity late pregnancy, within 6 hours delivery first piglet, on four commercial piglet-producing farms Finland. Microbial analyzed by 16S rRNA gene amplicon sequencing. most abundant genera oral Rothia , Moraxella Streptococcus . was dominated Clostridium sensu stricto 1 genus vagina colostrum. Moderate differences diversity observed between farms. relative abundances Neisseria (MaAsLin 2 q = 0.002, ANCOMBC 0.005), Fusobacterium 0.008, 0.04) Bacteroides < 0.005, 0.06) lower samples Romboutsia 0.07, 0.05), Turicibacter 0.08, 0.02) Lachnospiraceae_UCG_007 0.1, 0.05) higher multiparous compared primiparous In vaginal there a tendency than Among differentially taxa, F necrophorum nucleatum identified samples, gastrosuis S dysgalactiae samples. Twenty-eight samples were analysed 16 S metabarcoding either directly raw (raw microbiota, RM) or complete obtained from seven non-selective media (cultivable CM). approach enabled more in-depth investigation CM than conventional methods based individual sequencing subset isolates resulted moderate gain diversity within each sample. It confirmed that was lower RM, with ~ 7 versus 69 per sample RM respectively, an under-representation strictly anaerobic CM. In vitro gastrointestinal digestion overall good survival 6 but partial loss monocyte THP1 cell line, except Staphylococcus epidermidis strain gained potential. These results highlight to survive during interact intestinal epithelium immune system, well importance considering process when evaluating host-bacteria interactions.

In order to optimize diets for Bengal tiger cubs and improve their health condition survival rates, we conducted microbiota metabolomics analyses on fecal samples from fed goat dog milk replacer formulae. The results showed that there were significant differences in microorganisms metabolites between the two groups. At phylum level, major components of microbial composition feces Firmicutes, Actinobacteriota, Proteobacteria, Bacteroidota Fusobacteriota. addition, abundance gut varied significantly groups cubs. powder exhibited an increase probiotic bacteria (Anaerostipes Clostridium_scindens) (p < 0.05), community tended be more balanced. Metabolomics data further elucidated feeding different formulae affected metabolic pathways group, 76 up-regulated 278 down-regulated particularly affecting metabolism vitamin D3, B5, isoleucine, valine, phenylalanine oleic acid. same time, 19 including amino acid metabolism, lipid nucleotide pathways. conclusion, this study confirms formula affects These findings may provide new insights into how dietary strategies influence regulation overall

Human breast milk (HBM) is considered the gold standard for infant nutrition due to its optimal nutrient profile and complex composition of cellular non-cellular components. Breastfeeding positively influences newborn's gut microbiota health, reducing risk conditions like gastrointestinal infections chronic diseases (e.g., allergies, asthma, diabetes, obesity). Research has revealed that HBM contains beneficial microbes aid maturation through mechanisms antimicrobial production pathogen exclusion. The can be affected by several factors, including gestational age, delivery mode, medical treatments, lactation stage, as well maternal lifestyle habits diet, physical activity, sleep quality, smoking, alcohol consumption, stress level). Particularly, factors play a significant role in shaping directly modulating microbial or influencing enteromammary pathway. This narrative review current findings summarized how microbiota.
