Background Obesity is a chronic metabolic disease, mainly caused by excessive/abnormal fat accumulation, as well accompanied endotoxemia and inflammation. Quercetin, natural flavonoid, may alleviate obesity regulating gut microbiota metabolites, but its exact mechanism for improving unknown. Objectives The aim of this study was to investigate the effects quercetin on high-fat diet (HFD)-induced in mice. In particular, we focused regulatory tryptophan metabolite indole-3-propionic acid (IPA). Methods C57BL/6J mice were subjected 20-week HFD feeding regimen with concurrent daily oral administration or IPA. body weight, barrier function, inflammation determined. Gut composition analyzed 16S rRNA sequencing IPA levels measured serum feces. vitro experiments, Caco-2 cells used evaluate fecal dilutions from quercetin-treated tight junction protein expression aryl hydrocarbon receptor (AhR) activation. Results Our results revealed that supplementation significantly mitigated inflammation, improved disrupted function through actvation AhR/interleukin 22 (IL-22) pathway. treatment increased abundance Lactobacillus . Quercetin intervention feces alleviated enhanced intestinal AhR findings further corroborated cell experiment, which indicated modulation dysregulated change microbial coordinated improvement effect disruption. Conclusion alleviates restoring induced disorder, elevates level activate AhR/IL-22 pathway, thereby enhancing integrity suppressing
