Heat stress (HS) is a significant factor that adversely affects the health, welfare, and productivity of domestic animals, particularly impacting embryo implantation rates. However, effects HS on endometrial function during peri-implantation period in Hainan black goats remain unclear. This study explores influence endometrium these goats. We collected uterine tissue samples from both control heat-stressed performed vitro experiments using 2 × factorial design. design included two temperature conditions (37 °C as 42 to simulate heat stress) pharmacological treatments: chloroquine (CQ), an autophagy inhibitor, rapamycin (RAPA), activator. Our results showed initially suppresses activity, which subsequently enhanced with prolonged exposure. The pharmacologic modulation autophagy, through activation or inhibition, resulted corresponding upregulation downregulation epithelial cells' (EECs) receptivity markers. overexpression ATG7 partially reversed HS-induced Additionally, TJP1, tight-junction marker, was degraded under genetic but accumulated more EECs pre-treated CQ. These findings suggest plays protective role maintaining offers valuable insights into proposes potential strategy mitigate adverse goat reproduction.
