SARS-CoV-2 is the cause of a pandemic with growing global mortality. Using comprehensive identification RNA-binding proteins by mass spectrometry (ChIRP-MS), we identified 309 host that bind RNA during active infection. Integration this data ChIRP-MS from three other viruses defined viral specificity RNA-host protein interactions. Targeted CRISPR screens revealed majority functional protect virus-induced cell death, and comparative across seven shared SARS-specific antiviral factors. Finally, combining RNA-centric approach screens, demonstrated physical connection between mitochondria, highlighting organelle as general platform for activity. Altogether, these provide catalog interactions, which may inform studies to understand host-virus interface nominate pathways could be targeted therapeutic benefit.

During the coronavirus disease-2019 (COVID-19) pandemic, severe acute respiratory syndrome-related coronavirus-2 (SARS-CoV-2) has led to infection of millions people and claimed hundreds thousands lives. The entry virus into cells depends on receptor-binding domain (RBD) spike (S) protein SARS-CoV-2. Although there is currently no vaccine, it likely that antibodies will be essential for protection. However, little known about human antibody response SARS-CoV-21–5. Here we report 149 COVID-19-convalescent individuals. Plasma samples collected an average 39 days after onset symptoms had variable half-maximal pseudovirus neutralizing titres; titres were less than 50 in 33% samples, below 1,000 79% only 1% above 5,000. Neutralizing antibody responses to coronaviruses mainly target the receptor-binding domain (RBD) of trimeric spike. Here, we characterized polyclonal immunoglobulin Gs (IgGs) and Fabs from COVID-19 convalescent individuals for recognition coronavirus spikes. Plasma IgGs differed in their focus on RBD epitopes, alpha- beta-coronaviruses, contributions avidity increased binding/neutralization over Fabs. Using electron microscopy, examined specificities plasma Fabs, revealing both S1A epitopes SARS-CoV-2 Moreover, a 3.4 Å cryo-electron microscopy (cryo-EM) structure neutralizing monoclonal Fab-spike complex revealed an epitope that blocks ACE2 receptor binding. Modeling based these structures suggested different potentials inter-spike crosslinking by viruses, would not be affected identified spike mutations. Overall, our studies structurally define recurrent anti-SARS-CoV-2 class derived VH3-53/VH3-66 similarity SARS-CoV VH3-30 antibody, providing criteria evaluating vaccine-elicited antibodies.
