Abstract Lysine lactylation (Kla) is a newly discovered posttranslational modification that involved in important life activities, such as glycolysis-related cell function, macrophage polarization and nervous system regulation, has received widespread attention due to the Warburg effect tumor cells. In this work, we first design natural language processing method automatically extract 3D structural features of Kla sites, avoiding potential biases caused by manually designed features. Then, establish two prediction frameworks, Attention-based feature fusion model (ABFF-Kla) EBFF-Kla, integrate sequence structure based on layer embedding layer, respectively. The results indicate ABFF-Kla Embedding-based (EBFF-Kla), which fuse from protein sequences spatial structures, have better predictive performance than models use only Our work provides an approach for automatic extraction features, well flexible framework prediction. source code training data EBFF-Kla are publicly deposited at: https://github.com/ispotato/Lactylation_model.

Abstract For decades, lactate has been considered a byproduct of glycolysis. The shuttle hypothesis shifted the paradigm, demonstrating that not only plays important roles in cellular metabolism but also communications, which can transcend compartment barriers and occur within among different cells, tissues organs. Recently, discovery induce novel post-translational modification, named lysine lactylation (Kla), brings forth new avenue to study nonmetabolic functions for lactate, inspired ‘gold rush’ academic commercial interest. Abstract Histone post-translational modifications (HPTMs), as one of the core mechanisms epigenetic regulation, are garnering increasing attention due to their close association with onset and progression diseases potential targeted therapeutic agents. Advances in high-throughput molecular tools abundance bioinformatics data have led discovery novel HPTMs which similarly affect gene expression, metabolism, chromatin structure. Furthermore, a growing body research has demonstrated that histone also play crucial roles development various diseases, including cancers, cardiovascular infectious psychiatric disorders, reproductive system diseases. As a major product of glycolysis and vital signaling molecule, many studies have reported the key role lactate in tumor progression cell fate determination. Lactylation is newly discovered post-translational modification induced by lactate.
