Abstract
Lysine
lactylation
(Kla)
is
a
newly
discovered
posttranslational
modification
that
involved
in
important
life
activities,
such
as
glycolysis-related
cell
function,
macrophage
polarization
and
nervous
system
regulation,
has
received
widespread
attention
due
to
the
Warburg
effect
tumor
cells.
In
this
work,
we
first
design
natural
language
processing
method
automatically
extract
3D
structural
features
of
Kla
sites,
avoiding
potential
biases
caused
by
manually
designed
features.
Then,
establish
two
prediction
frameworks,
Attention-based
feature
fusion
model
(ABFF-Kla)
EBFF-Kla,
integrate
sequence
structure
based
on
layer
embedding
layer,
respectively.
The
results
indicate
ABFF-Kla
Embedding-based
(EBFF-Kla),
which
fuse
from
protein
sequences
spatial
structures,
have
better
predictive
performance
than
models
use
only
Our
work
provides
an
approach
for
automatic
extraction
features,
well
flexible
framework
prediction.

Abstract
For
decades,
lactate
has
been
considered
a
byproduct
of
glycolysis.
The
shuttle
hypothesis
shifted
the
paradigm,
demonstrating
that
not
only
plays
important
roles
in
cellular
metabolism
but
also
communications,
which
can
transcend
compartment
barriers
and
occur
within
among
different
cells,
tissues
organs.
Recently,
discovery
induce
novel
post-translational
modification,
named
lysine
lactylation
(Kla),
brings
forth
new
avenue
to
study
nonmetabolic
functions
for
lactate,
inspired
‘gold
rush’
academic
commercial
interest.
Zhang
et
al.
first
showed
Kla
is
manifested
histones
as
epigenetic
marks,
then
mounting
evidences
demonstrated
occurs
diverse
non-histone
proteins.
widespread
faithfully
orchestrates
numerous
biological
processes,
such
transcription,
inflammatory
responses.
Notably,
dysregulation

myriad
pathological
processes.
In
this
review,
we
comprehensively
reviewed
curated
existing
literature
retrieve
identified
sites
on
both
proteins
summarized
recent
major
advances
toward
its
regulatory
mechanism.
We
thoroughly
investigated

underlying
signaling
pathway
summarize
how
regulates
various
processes
normal
physiological
states.
addition,
further
highlight
effects
development
human
diseases
including
inflammation
response,
tumorigenesis,
cardiovascular
nervous
system

complex
diseases,
might
potentially
contribute
deeply
understanding

Abstract
Histone
post-translational
modifications
(HPTMs),
as
one
of
the
core
mechanisms
epigenetic
regulation,
are
garnering
increasing
attention
due
to
their
close
association
with
onset
and
progression
diseases
potential
targeted
therapeutic
agents.
Advances
in
high-throughput
molecular
tools
abundance
bioinformatics
data
have
led
discovery
novel
HPTMs
which
similarly
affect
gene
expression,
metabolism,
chromatin
structure.
Furthermore,
a
growing
body
research
has
demonstrated
that
histone
also
play
crucial
roles
development
various
diseases,
including
cancers,
cardiovascular
infectious
psychiatric
disorders,
reproductive
system
diseases.
This
review
defines
nine
modifications:
lactylation,
citrullination,
crotonylation,
succinylation,
SUMOylation,
propionylation,
butyrylation,
2-hydroxyisobutyrylation,
2-hydroxybutyrylation.
It
comprehensively
introduces
modification
processes
these
HPTMs,
transcription,
replication,
DNA
repair
recombination,
structure,
well
involvement
promoting
occurrence
clinical
applications
targets
biomarkers.
Moreover,
this
provides
detailed
overview
HPTM
inhibitors
targeting
emerging
strategies
treatment
multiple
while
offering
insights
into
future
prospects
challenges.

ABSTRACT
In
recent
years,
a
