The interaction between the tumor microenvironment (TME) and cancer cells is a complex mutually beneficial system that leads to rapid proliferation, metastasis, resistance therapy. It now recognized are not isolated, progression governed among others, by many components of TME. reciprocal cross-talk their can be indirect through secretion extracellular matrix (ECM) proteins paracrine signaling exosomes, cytokines, growth factors, or direct cell-to-cell contact mediated cell surface receptors adhesion molecules. Among TME components, cancer-associated fibroblasts (CAFs) unique interest. As one most abundant TME, CAFs play key roles in reorganization matrix, facilitating metastasis chemotherapy evasion. Both have been described for modulating progression. In this review, we focus on recent advances understanding role driving development metastasis. We also summarize findings resistance.

Tumor-derived extracellular vesicles (EVs) play an important role in cancer progression. Neutral sphingomyelinases (nSMases) are lipid-modifying enzymes that modulate the secretion of EVs from cells. How nSMase activity and therefore ceramide generation affect composition functionality secreted is not fully understood. Here, we aimed to investigate expression nSMases 1 2 prostate (PCa) tissue their EV for cell migration. Reduced was found correlated with age patient. When inhibited by GW4869 PCa cells (PC3 DU145), secretome significantly altered, while number total protein content released were changed. Using proteomic analysis, matrix proteins, such as SDC4 (Syndecan-4) SRPX-2, differentially on GW4869-treated PC3 In scratch wound migration assays, increased compared control but DU145 cells, knockdown reduced These other nSMase-2-dependent proteins interesting candidates understanding stress-induced progression cancer.
