During coevolution, plants produce numerous toxic secondary metabolites to protect themselves. However, most still serve as food for insects meet their nutritional needs. The evolutionary processes that enable herbivorous resist plant defenses remain largely complex and difficult predict. In this study, lactase phlorizin hydrolase (LPH) was identified the first time in Aphis gossypii. Bioinformatics analysis showed LPH protein belongs glycoside 1 (GH1) family. qPCR transcript level of cotton aphids treated with expression dsAgCYP6CY3 lines significantly reduced by 39.9% at 48 h compared nontransgenic cotton, hydrolytic activities on lactose were 28.7% 20.1%, respectively. vitro enzyme activity experiments demonstrated could hydrolyze four phenolic glycosides. addition, RNA interference (RNAi) insect performance assays silencing affected growth larvae, resulting death larvae exposed These results reveal an scenario whereby herbivores use deglycosylation develop resistance can be exploited protection.

The ecdysone-induced transcription factor E93 in model insects plays multiple roles the insect metamorphosis processes, such as remodeling larval tissues and determining adult tissue formation. knockdown of leads to incomplete metamorphosis, suggesting that is a potential target for pest control. In this study, HaE93 gene cotton bollworm Helicoverpa armigera, polyphagous various commercial crops worldwide, was identified found have high expression egg, prepupal, pupal stages. injection dsHaE93 induced about 60% mortality H. armigera at larval-pupal stage. About 30% surviving showed delayed pupation developed abnormal wings, females reduced ovaries. 90% individuals failed reproduce before they died. results qRT-PCR levels ecdysone primary-response genes, chitin synthesis-related wing ovary development-related genes were armigera. These indicated critical role larva-pupa-adult development cuticle, wing, female by regulating associated genes. Bioassays administered either oral delivery or similar results, which suggested can be used RNAi control

MicroRNAs (miRNAs) are post-transcriptional gene regulators. In the miRNA pathway's cytoplasmic part, is processed from a hairpin-structured precursor to double-stranded (ds) mature RNA and ultimately single-stranded miRNA. insects, ingesting these two ds forms can regulate target expression; this inspired trophic miRNA's use as functional genomics pest management tool. However, systematic studies enabling comparisons of pre- forms, dosages, administration times instar-wise effects on transcripts phenotypes, which help develop method, unavailable due different focuses previous investigations. We investigated impact trophically delivered Px-let-7 lepidopteran Plutella xylostella, compare efficacies its ds-mature forms. Continuous feeding miRNA-supplemented diet suppressed expressions FTZ-F1 E74, ecdysone pathway genes. Both pre-let-7 let-7 similarly downregulated in all four larval instars. Pre-let-7 ingestions decreased mass instar duration increased mortality instars, exhibiting adverse growth development. processing Dicer-1 AGO-1's upregulations upon ingestion denoted systemic spread tissues. The scrambled sequence controls did not affect transcripts, suggesting sequence-specific targeting by hairpin cassette's non-involvement downregulation. This work provides framework for function analyses potentiates utility management.
