Abstract BACKGROUND Laccase2 is a member of the multicopper oxidase family that plays essential roles in sclerotization and pigmentation insect cuticle. Previous studies have suggested RNA interference (RNAi) targeting laccase2 often resulted high mortality various pests. This study aims to evaluate potential effects gene knockdown on serious pest, Halyomorpha halys (Stål) (Hemiptera: Pentatomidae). RESULTS We identified, validated characterized one ( Hhlac2 ) from H. . Quantitative real‐time PCR results demonstrated expression tissue‐specific changes periodically during development. The revealed injecting nymphs with double‐stranded (ds at doses ranging 100 500 ng/individual significantly suppressed expression, was not inhibited until injection dose reached more than ng/individual, while rate over 90% ng. RNA‐sequencing analysis 802 up‐regulated 1052 down‐regulated genes ds ‐injected group compared control, these unigenes were mainly enriched terms tyrosine metabolism, melanin biosynthesis, chitin, immunity apoptosis. Notably, several key differentially expressed associated metabolism (tyrosine lipid chitin metabolism) innate (Toll signaling pathway) suppressed, which may influence host fitness together phenotypic increased mortality. CONCLUSION Our elucidates pivotal induces overall disorder molecular individual levels, respectively, provide insight into biocontrol via RNAi‐based technology heteropteran © 2025 Society Chemical Industry.
