Background: Pathological progression from non-alcoholic fatty liver disease (NAFLD) to fibrosis (LF) hepatocellular carcinoma (HCC) is a common dynamic state in many patients. Curcumin, dietary supplement derived the turmeric family, expected specifically inhibit development of this progression. However, there lack convincing evidence. Methods: The studies published until June 2023 were searched PubMed, Web Science, Embase, and Cochrane Library databases. SYstematic Review Center for Laboratory animal Experimentation (SYRCLE) approach was used evaluate certainty StataSE (version 15.1) Origin 2021 software programs analyze critical indicators. Results: Fifty-two involving 792 animals included, three models reported. Curcumin demonstrates significant improvement key indicators across stages NAFLD, fibrosis, HCC. We conducted detailed analysis inflammatory markers IL-1β, IL-6, TNF-α, which traverse entire process. research results reveal that curcumin effectively hinders at each stage by suppressing inflammation. exerted hepatoprotective effects dose range 100 400 mg/kg treatment duration 4 10 weeks. mechanistic reveals primarily exerts its modulating multiple signaling pathways, including TLR4/NF-κB, Keap1/Nrf2, Bax/Bcl-2/Caspase 3, TGF-β/Smad3. Conclusion: In summary, has shown promising therapeutic during overall NAFLD–LF–HCC. It inhibited pathological synergistic mechanisms related anti-inflammatory, antioxidant, apoptosis regulation.

BackgroundTraditional Chinese Medicine (TCM), has been used for hepatocellular carcinoma (HCC) at every therapeutic stage, even before tumor formation. However, the efficacy of TCM in reducing incidence HCC patients with chronic hepatitis B-related cirrhosis remains unclear. This study aims to address this gap.MethodsPublications were collected from PubMed, EMBASE, Cochrane Library, Web Science, CNKI, Sino Med, VIP, and Wan Fang Databases. Relative risk (RR) was calculated a 95 % confidence interval (CI). Heterogeneity assessed. The Collaboration's tool assess bias.Results10 studies 2702 showed that combination therapy significantly reduced post-hepatitis B 1, 3, 5 years. preventive effects compensated cirrhosis, but not decompensated cirrhosis. Furthermore, correlated improved liver function enhanced virological response.ConclusionCombination demonstrated certain potential is attrinuted improvement enhancement viral response. field still needs more high-quality RCTs provide stronger evidence future.

Liver cancer is one of the most prevalent cancers worldwide. The first-line therapeutic drug sorafenib offers only a moderate improvement in patients' conditions. Therefore, an approach to enhancing its efficacy urgently needed. It has been revealed that hepatocellular carcinoma (HCC) cells with heightened intracellular quiescin sulfhydryl oxidase 1 (QSOX1) exhibit increased sensitivity sorafenib. QSOX1 secreted disulfide catalyst, and it widely recognized extracellular promotes growth, invasion, metastasis through participation establishment matrix. Inhibiting secretion can increase decrease QSOX1. Such would sensitize HCC but remains be established. Since (-)-epigallocatechin-3-gallate (EGCG) demonstrated effective inhibitor α-fetal protein from cells, we screened inhibition using polyphenolic compounds. We examined eight dietary polyphenols (EGCG, quercetin, fisetin, myricetin, caffeic acid, chlorogenic resveratrol, theaflavin) found EGCG quercetin effectively inhibited human (HepG2 or Huh7), resulting high low combination both which change cellular distribution QSOX1, sorafenib, no influence on exhibited multiple synergistic effects against including induction apoptosis invasion metastasis. In conclusion, our current results suggest have potential developed as adjuvants treatment by modulating

Scope Type 2 diabetes and nonalcoholic fatty liver diseases (NAFLDs) are promoted by insulin resistance (IR), which alters lipid homeostasis in the liver. This study aims to investigate effect of N ‐methylpyridinium (NMP), a bioactive alkaloid coffee brew, on metabolism hepatocytes. Methods results The NMP modulating is evaluated at physiological concentrations cell model represented HepG2 cells cultured high‐glucose medium. Hyperglycemia triggers droplet accumulation enhances lipogenic gene expression, transactivated sterol regulatory element binding protein‐1 (SREBP‐1). Lipid redox status endoplasmic reticulum (ER) stress, leading activation unfolded protein response antioxidative pathways X‐Box Binding Protein 1(XBP‐1)/eukaryotic Initiation Factor alpha (eIF2α) Kinase RNA‐Like ER nuclear factor erythroid 2‐related (NRF2), respectively. induces phosphorylation AMP‐dependent kinase (AMPK) acetyl‐CoA carboxylase α (ACACA), improves homeostasis, essential steps reduce lipogenesis accumulation. Conclusion These suggest that may be beneficial for management T2D NAFLD ameliorating oxidative metabolism.

Nonalcoholic fatty liver disease (NAFLD), represents a chronic progressive that imposes significant burden on patients and the healthcare system. Linggui Zhugan decoction (LGZGD) plays substantial role in treating NAFLD, but its exact molecular mechanism is unknown. Using network pharmacology, this study aimed to investigate of action LGZGD NAFLD. Active ingredients targets were identified through integration data from TCMSP, GEO, GeneCards, OMIM databases. Cytoscape 3.9.1 software, conjunction with STRING platform, was employed construct diagrams screen core targets. The enrichment analysis gene ontology Kyoto Encyclopedia Genes Genomes pathways conducted by using R. Molecular docking active performed AutoDock Vina software. We obtained 93 112 potential bioinformatic primary included quercetin, kaempferol, naringenin. AKT1, MYC, HSP90AA1, HIF1A, ESR1, TP53, STAT3. Gene function revealed associations responses nutrient oxygen levels, nuclear receptor activity, ligand-activated transcription factor activity. signaling pathway implicated involvement PI3K-Akt, IL-17, TNF, Th17 cell differentiation, HIF-1, TLR pathways. studies indicated strong binding affinities between intervenes NAFLD multi-ingredient, multi-target, multi-pathway approach. Treatment can improve insulin resistance, oxidative stress, inflammation, lipid metabolism associated
