Cutaneous melanoma (CM) is the most aggressive and fatal malignancy among other skin cancers its incidence has risen steadily recently around world. Hormone-related therapy, particularly estrogen (E2) been used as a prospective strategy for CM treatment. Quercetin luteolin are flavonoids with antitumor effects against wide range of including CM. However, underlying mechanism their actions through GPER in not fully understood. We examined anti-tumor quercetin on A375 cell line activation G-protein coupled receptor (GPER). MTT assay was performed to assess impact viability. Apoptosis cycle were studied by flow cytometry. Cell migration evaluated transwell assay. expression effect key signaling proteins confirmed immunofluorescence staining Western blot, respectively. Results showed that inhibited proliferation migration, induced apoptosis, blocked at S G2/M cells. Immunofluorescence immunoblotting data demonstrated presence this two enhanced except high concentration 100 µM. P-ERK c-Myc expression, an abolished antagonist G15, confirming GPER-mediated signaling. In conclusion, exhibited cells via activation, suggesting potential anti-melanoma therapeutics.
