Abstract SOBERANA ® 02 is a safe and effective conjugate vaccine against SARS-CoV-2, produced using the maleimide-thiol chemistry. In this vaccine, Cys 538 in recombinant receptor binding domain (RBD) of SARS-CoV-2 linked, through thiosuccinimide linker, to lysine residues tetanus toxoid (TT) preparation. LC-MS/MS analysis revealed that TT complex mixture proteins, similar other TTs where detoxified neurotoxin (d-TeNT) has been shown be most abundant protein (30-56%), regardless quantification method used. The fifteen proteins account for approximately 78% total proteins. activation process showed 102 out 107 d-TeNT incorporated maleimide group. contrast, when tryptic peptides isolated by Ni 2+ -NTA affinity chromatography, were analyzed LC-MS/MS, only 22 Lys cross-linked RBD C -terminal peptide ( CVNF 541 -HHHHHH), probably due steric hindrance. Twelve eighteen conjugation sites assigned based on identification linear carrying conjugated (Δm = +1454.58 Da or Δm +1472.59 Da) with stabilized linker forms, respectively.
