Antimicrobial phototherapy has gained recognition as a promising approach for addressing bacterial biofilms, however, its effectiveness is often impeded by the robust physical and chemical defenses of biofilms. Traditional antibacterial nanoplatforms face challenges in breaching extracellular polymeric substances barrier to efficiently deliver photosensitizers deep into Moreover, prevalent hypoxia within biofilms restricts success oxygen-reliant phototherapy. In this study, we engineered soft mesoporous organosilica nanoplatform (SMONs) incorporating polyethylene glycol (PEG), catalase (CAT), indocyanine green (ICG), forming SMONs-PEG-CAT-ICG (SPCI). We compared antimicrobial efficacy SPCI with more rigid nanoplatforms. Our results demonstrated that unique flexible mechanical properties enable it navigate through biofilm barriers, markedly enhancing ICG penetration methicillin-resistant Staphylococcus aureus (MRSA) Notably, murine subcutaneous MRSA infection model, showed superior pharmacokinetic benefits over counterparts. Macrophages and osteocytes are important regulators of inflammation, osteogenesis osteoclastogenesis. However, their interactions under adverse conditions, such as biomaterial-associated infection (BAI) not fully understood. We aimed to elucidate how factors released from macrophages modulate osteocyte responses in an vitro indirect 3D co-culture model. Human monocyte-derived were cultured on etched titanium disks activated with either IL-4 cytokine (anti-inflammatory M2 phenotype) or Staphylococcus aureus secreted virulence simulate BAI (pro-inflammatory M1 phenotype). Primary collagen gels then stimulated conditioned media (CM) these macrophages. The response was analyzed by gene expression, protein secretion, immunostaining. phenotype confirmed IL-1β TNF-α ARG-1 MRC-1.Osteocytes receiving CM revealed bone inhibitory effects, denoted reduced secretion formation osteocalcin (BGLAP) increased the sclerostin (SOST). These also downregulated pro-mineralization PHEX upregulated anti-mineralization MEPE. Additionally, exhibited pro-osteoclastic potential upregulating RANKL expression. Nonetheless, M1-stimulated expressed a higher level potent pro-osteogenic factor BMP-2 parallel downregulation inhibitor genes DKK1 SOST, suggesting compensatory feedback mechanisms. Conversely, M2-stimulated mainly anti-osteoclastic OPG anti-catabolic effect. Altogether, our findings demonstrate strong communication between (BAI)-simulated that effects osteoblastic osteoclastic processes partly mediated via this communication. Biomaterial-associated infections major challenges underlying mechanisms cellular missing, especially among cells inflammatory side (macrophages key cell bacterial clearance) regenerative (osteocyte main regulator bone). evaluated effect macrophage polarization driven stimulation function using model, hence mimicking scenario infection. results suggest at least part biomaterial associated macrophage-to-osteocyte

Abstract Bacterial plant diseases, worsened by biofilm‐mediated resistance, are increasingly threatening global food security. Numerous attempts have been made to develop agrochemicals that inhibit biofilms, however, their ineffective foliar deposition and difficulty in removing mature biofilms remain major challenges. Herein, multifunctional three‐component supramolecular nano‐biscuits (NI6R@CB[7]@ β ‐CD) successfully engineered via ordered self‐assembly between two macrocycles [cucurbit[7]uril (CB[7]), ‐cyclodextrin ( ‐CD)] R )‐2‐naphthol‐based bis‐imidazolium bromide salt (NI6R). This macrocycles‐involved bactericidal material combines many advantages. 1) Alleviate the off‐target movement of droplets on hydrophobic blade surfaces. 2) Enhance biofilm‐disrupting ability. At a low‐dose 4.44 µg mL −1 , inhibition rate biofilm formation reached 78.3%. 35.5 potency remove 77.6%. 3) Efficiently hinder bacterial reproduction, swimming, extracellular polysaccharide production, enzyme secretion, virulence plants. These superior characteristics undoubtedly transmitted vivo control effect. 200 this smart exhibits efficiencies 49.6%/65.0%/85.4% against three kinds diseases (rice leaf blight, citrus canker, kiwifruit canker), respectively, surpassing commercial bactericide—thiodiazole‐copper‐20%SC (33.6%/41.5%/43.2%) NI6R (40.3%/51.2%/71.2%). Furthermore, NI6R@CB[7]@ ‐CD is biosafe non‐target organisms. study instructive for constructing sustainable crop protection.
