Dephosphorylation is an essential process in cellular signaling with protein phosphatases playing a critical role functions and disease mechanisms. Herein, novel ultrasensitive photoelectrochemical (PEC) biosensing platform for detecting tyrosine phosphatase 1B (PTP1B) activity was developed, based on the sensitization effect of magnetic ZnFe2O4@ZrMOF nanoenzyme integrated two-step radical polymerization signal amplification strategy. The PTP1B-specific phosphorylated peptide (p-peptide) immobilized 96-well plate then coupled to through coordination between its phosphate groups Zr4+ ions. When PTP1B present, p-peptide specifically recognized dephosphorylated, causing release ZnFe2O4@ZrMOF. After separation, detached nanoenzyme, peroxidase (POD)-like photoresponsive oxidase (OXD)-like activities, used as probe, which not only exhibited PEC AgInS2/Ag2S/magnetic ITO (MITO) photoelectrode but also triggered reactions form ZnFe2O4@ZrMOF/polydopamine (PDA)/ poly(ferrocenylmethyl methacrylate) (PFcMMA) composite further enhancing amplification. proposed achieved detection activity, demonstrating wide linear range (10 aM-0.1 μM) ultralow limit (3.92 aM), along good reproducibility, satisfactory stability, high selectivity, promising practical applicability. This work provides prospective method assay, facilitating early diagnosis therapeutic development alongside innovative strategy advancing biosensor sensitivity.
