Isoorientin (ISO) is a natural lignan glycoside flavonoid found in various plants, including Charcot and Stonecrop. ISO exhibits diverse physiological pharmacological effects, such as antioxidative, anti-inflammatory, hepatoprotective, antiviral, antianxiety, anti-myocardial ischaemic properties, well lipid metabolism regulation. This study investigated the impact of supplementation on oxidative stress accumulation porcine early embryos, along with its underlying mechanisms. Porcine embryos were cultured vitro under different concentrations (0, 1, 10, 100 nM). The results revealed that 10 nM significantly enhanced blastocyst rate total embryonic cell count vitro. ISO-treated exhibited reduced reactive oxygen species levels elevated glutathione compared to untreated group. In addition, treatment increased expression key antioxidant regulator Nrf2, improved mitochondrial function, droplet accumulation. Concurrently, embryo autophagy apoptosis decreased. Furthermore, upregulated antioxidant-related genes (SOD1, SOD2, CAT) biogenesis related (NRF1, NRF2, SIRT1), while downregulating synthesis-related (SREBP1 FASN). Additionally, hydrolysis-related (ACADS) elevated. These findings collectively suggest may facilitate development pigs by ameliorating metabolism.

Abstract Imidacloprid (IMI) is a neonicotinoid insecticide with the highest global market share, and IMI exposure in environment can negatively affect many nontarget organisms (a general term for affected by drugs other than target organisms). Resveratrol (RSV), non‐flavonoid polyphenolic organic compound derived from peanuts, grapes, plants, has anti‐inflammatory antioxidant effects. It currently unclear how RSV protects against cell damage caused IMI. Therefore, we established an experimental model of chicken lymphocyte lines exposed to 110 μg/mL and/or 0.5 μM 24 h. According results, markedly raised intracellular reactive oxygen species levels diminished activity cellular enzymes (CAT, SOD, GPx), leading MDA accumulation decreased T‐AOC. JNK , ERK P38 essential components mitogen‐activated protein kinase (MAPK) signaling pathway, were also expressed more when was present. Additionally, resulted upregulation mitochondrial apoptosis ( Caspase 3 9 Bax Cyt‐c ) necroptosis 8 RIPK1 RIPK3 MLKL related factors expression, downregulation Bcl‐2 induction cytokine IL‐6 TNF‐α IFN‐γ expression. The combined treatment significantly reduced oxidative stress levels, inhibited MAPK alleviated IMI‐induced apoptosis, necroptosis, immune dysfunction. To summarize, antagonized dysfunction inhibiting ROS/MAPK pathway.

Chrysoeriol (CHE) is a flavonoid substance that exists in many plants. It has various physiological and pharmacological effects, including anti-inflammatory, antioxidant, anti-tumor, protective activity, especially for the cardiovascular system liver. Among common livestock embryos, porcine embryos are often considered high-quality objects studying antioxidant mechanisms of oocytes. Because contain high levels lipids, they more vulnerable to external stimuli, which affect development. Our study explored influence CHE supplementation on oxidative stress oocytes its possible mechanisms. Different concentrations (0, 0.1, 1, 3 µM) were supplemented vitro culture medium The results showed with 1 µM significantly increased blastocyst rate total cell number vitro. After finding beneficial effects CHE, we measured reactive oxygen species (ROS), glutathione (GSH), mitochondrial membrane potential (MMP) when reached 4-cell stage development determined apoptosis, proliferation, autophagy at expression some related genes preliminarily detected by qRT-PCR. apoptosis blastocysts CHE-treated also decreased compared untreated culture. Furthermore, downregulated intracellular ROS GSH embryos. was shown improve activity mitochondria inhibit occurrence autophagy. In addition, antioxidant-related (SOD1, SOD2, CAT) pluripotency-related (SOX2, OCT4, NANOG) upregulated. At same time, apoptosis-related (Caspase 3) autophagy-related (LC3B) downward trend after CHE. These indicate improved reducing levels.

Ageing is a natural process characterized by time-dependent decline of physiological integrity that compromises functionality and inevitably leads to death. This also quite relevant in major human pathologies, being primary risk factor neurodegenerative diseases, metabolic disorders, cardiovascular diseases musculoskeletal disorders. Bearing this mind, it not surprising research aiming at improving health during has burst the last decades. Importantly, hallmarks ageing phenotype have been identified, knowledge for future studies towards identification putative pharmaceutical targets, enabling development preventive/therapeutic strategies improve longevity. In context, aromatic plants emerged as source potential bioactive volatile molecules, mainly monoterpenes, with many referring their anti-ageing potential. Nevertheless, an integrated review on current lacking, several approaches studying isolated or overall effect, without depicting possible mechanisms action. Herein, we aim provide updated systematization monoterpenes recently proposed hallmarks, highlight main action already well chemical entity–activity relations. By gathering categorizing available scattered information, identify important gaps could help pave way field.
