The aim of the current work was to examine for first time nephropreventive capacity Ephedra alata seed extract (E) against maternal exposure acephate in rat offspring. vivo results revealed that E. supplementation 28 days (40 mg/kg b.w.) significantly attenuated nephrotoxicity adult offspring induced by acephate. In fact, it decreased levels creatinine and uric acid increased albumin content compared intoxicated group. utero studies showed inhibited renal oxidative stress generated reducing lipid peroxidation enhancing antioxidant biomarker activities (GSH, CAT, SOD). inhibition DNA fragmentation improvement ultrastructural changes highlighted prophylactic effect tissue. Additionally, immunofluorescence study upregulation LC3 gene expression, suggesting stimulate autophagic processes as a protective mechanism. Molecular docking analysis indicated hexadecasphinganine, major compound alata, has higher affinity toward Na+/K+-ATPase, epithelial sodium channel (ENaC), hydrogen exchanger 3 (NHE3) genes than Hexadecasphinganine could be considered potential inhibitor activity these therefore exerted its preventive capacity. obtained findings confirmed capacities, which related bioactive compounds, possess activities.

The adverse effects of tetramethrin residues, a synthetic pyrethroid used in many insecticide formulations, on environmental health and living organisms are matter concern. aim this study was to evaluate the genotoxic developmental non-target organism, Drosophila melanogaster. Abstract Background Septic acute kidney injury (SAKI) is usually caused by sepsis. It has been shown that catalpol (Cat) impairs sepsis‐evoked organ dysfunction to a certain degree. The current work aims evaluate the protective effects of Cat on SAKI and potential mechanisms in vivo vitro. Methods cellular murine model were set up using lipopolysaccharide (LPS) vitro vivo. Cell apoptosis cells was determined TUNEL assay. Levels inflammatory cytokines detected enzyme‐linked immunosorbent assay (ELISA). levels markers oxidative evaluated corresponding commercial kits. Protein assayed via western blotting immunohistochemistry (IHC) staining. Results results demonstrated LPS upregulated TNF‐α, IL‐6, malondialdehyde levels, downregulated superoxide dismutase, whereas treated have opposite results. Functional assays displayed remarkably reversed LPS‐challenged damage as impairment TNF‐α IL‐6 stress, HK‐2 cells. Moreover, knockdown Sirtuin 1 (Sirt1) counteracted suppressive impact LPS‐triggered response, renal damage. Further, elevated Sirt1 expression activated Nrf2/HO‐1 signaling LPS‐engendered Conclusion Our study clearly proved protected against LPS‐induced synergic antioxidant anti‐inflammatory actions regulating pathways.
