Mosquitoes, as major vectors of dangerous diseases, demand effective control measures for public health. Larvicides, while offering a potential solution, can exhibit varying effects on different mosquito species and developmental stages during efficacy studies. Abstract Background This study determined the blood-feeding patterns of wild-caught sandflies collected from provinces (Aydin and Mugla) in Aegean region Türkiye. Adult were using three CDC light traps per village August October 2024. A total 280 sandfly specimens (194 females 86 males) during study. Of 194 female specimens, 38 (23 Aydin 15 found to be blood-fed, 137 non-blood-fed, 19 gravid. In total, DNA was extracted blood-fed Invitrogen PureLink genomic isolation kit. To determine these insects, mitochondrial cytochrome b (cytb) gene amplified via multiplex PCR specific primers. Results The results showed that 23 sand fly sampled locality fed mostly cows (60.5%) followed by dogs (21%) chickens (18.4%). All samples blood whereas for Mugla locality, 53% 46% birds. Host preference also subjected ELISA reaction identify whose host could not see if there multiple preferences.
