Objective: Co-occurrence of schizophrenia/schizoaffective disorder (SSD) and stimulant use (StUD) is an ongoing clinical problem can lead to poor outcomes. Although emerging evidence has suggested psychostimulant substitution therapy may result in improved outcomes those with StUD, the efficacy safety for StUD concurrent SSD uncertain. This review aims systematically find assess all available on prescription psychostimulants co-occurring StUD. Methods: Electronic searches MEDLINE, PsycINFO, Embase, Scopus, ClinicalTrials, EU Clinical Trials, CADTH were conducted from inception February 27, 2024. Any study design was accepted if they involved following concepts 1) 2) psychostimulants. Given paucity trials meeting criteria, interest described qualitatively. Risk bias assessed using Q-Coh ROB2. Results: Only seven articles met most these case reports series. The single RCT included at high risk bias. Outcomes abstinence, reductions non-prescribed use, psychiatric hospitalizations, levels craving, improvements mental health, psychosocial functioning, adherence antipsychotic medications, retention treatment. Most results indicated that individuals SSD-StUD not associated Conclusion: Available treatment via lacking. More exploration required this question allow current practice be backed by evidence.

Background: To estimate the prevalence of co-occurring obsessive–compulsive disorder (OCD) with substance use (SUD) in general population. Methods: A comprehensive literature search was conducted on comorbid OCD and SUD population using MEDLINE, PsycINFO, EMBASE CINAHL. Using keywords, relevant studies published between 1993 2021 were identified. These analysed Preferred Reporting Items for Systematic Reviews Meta-Analysis (PRISMA) guidelines Meta-analysis Observational Studies Epidemiology. Results: Fourteen met inclusion criteria had a total combined sample size 47,850 participants. The lifetime any identified patients ranged 4.3% 62.4%. Among AUD, 14.1% 35.9%; cannabis OCD, 11.5% 24.4%; tobacco 15% 23.1%. Alcohol commonest psychoactive used most reviewed highest males. Conclusions: In conclusion, this review highlights variable SUDs individuals emphasizing that association differs based type substance. Overall, rate underscores need further research relationship these conditions development effective treatment strategies address both disorders concurrently.

While the majority of people with cocaine use disorders (CUD) also co-use tobacco/nicotine, most preclinical research does not include nicotine. The present study examined nicotine and under several conditions intravenous drug self-administration in monkeys, as well potential peripheral biomarkers associated co-use. In Experiment 1, male rhesus monkeys (N=3) self-administered (0.001-0.1 mg/kg/injection) alone (0.01-0.03 a progressive-ratio schedule reinforcement. When was added to cocaine, there significant leftward/upward shift number injections received. 2, socially housed female cynomolgus (N=14) concurrent drug-vs-food choice Adding solution shifted dose-response curves left, more robust shifts noted animals. There no evidence social rank differences. To assess reinforcing strength, delays were presentation drug; required significantly longer decrease choice, compared alone. Blood samples obtained post-session used analyze concentrations neuronally derived small extracellular vesicles (NDE); differences NDE profile observed for kappa-opioid receptors when co-used each controls. These results suggest that interactions involving are simply changing potency, but rather resulting changes strength should be utilized better understand neuropharmacology CUD evaluation treatments.

The global issue of substance abuse demands ongoing initiatives aligned with the United Nations Sustainable Development Goals. With drug use remaining prevalent worldwide, interventions are critical to addressing associated health challenges and societal implications. Exercise physical activities have emerged as integral components disorder (SUD) treatment, offering promising avenues for prevention, intervention, recovery. Recent research underscores efficacy exercise in reducing cravings, promoting abstinence, improving overall well-being. However, integrating into SUD recovery programs presents such dropout rates cultural considerations. This paper synthesizes existing literature on integration recovery, highlighting strategies enhancing treatment outcomes barriers adherence. Drawing cognitive–behavioral therapy, experiential learning, motivational interviewing, goal-setting techniques, holistic approach outlined this aims empower individuals both mentally physically, fostering resilience supporting long-term In conclusion, new need be taken by advocating inclusive policies, community engagement, collaborations across sectors. By doing so, stakeholders can optimize effectiveness contribute sustainable rehabilitation efforts SUD.

Fentanyl is a synthetic opioid widely used for its potent analgesic effects in chronic pain management and intraoperative anesthesia. However, high potency, low cost, accessibility have also made it significant drug of abuse, contributing to the global epidemic. This review aims provide an in-depth analysis fentanyl's medical applications, pharmacokinetics, metabolism, pharmacogenetics while examining adverse forensic implications. Special attention given misuse, polydrug interactions, challenges determining cause death fentanyl-related fatalities. misuse has escalated dramatically, driven by substitution heroin availability through online platforms, including dark web. Polydrug use, where fentanyl combined with substances like xylazine, alcohol, benzodiazepines, or cocaine, exacerbates toxicity increases risk fatal outcomes. undergoes rapid distribution, metabolism CYP3A4 into inactive metabolites, renal excretion. Genetic polymorphisms CYP3A4, OPRM1, ABCB1 significantly influence individual responses fentanyl, affecting efficacy potential toxicity. Fentanyl's side include respiratory depression, cardiac arrhythmias, gastrointestinal dysfunction, neurocognitive impairments. Chronic disrupts brain function, contributes mental health disorders, poses risks younger older populations alike. Fentanyl-related deaths require comprehensive investigations, judicial inspections, autopsies, toxicological analyses. Additionally, co-administration xylazine presents distinct scientific community. Histological immunohistochemical studies are essential understanding organ-specific damage, pharmacogenetic testing can identify susceptibilities. The growing prevalence abuse highlights need robust protocols, advanced research variability, strategies mitigate misuse. International collaboration, public education, harm reduction measures critical addressing crisis effectively.
